目的 探讨分泌性白细胞蛋白酶抑制因子( SLPI)在多囊卵巢综合征(PCOS)大鼠模型卵巢中的表达及生物学作用.方法 16只雌性Sprague-Dawley( SD)大鼠随机分为实验组和正常对照组(各8只).实验组以胰岛素联合人绒毛膜促性腺激素法建立SD大鼠的PCOS模型,正常对照组注射生理盐水.观察两组大鼠卵巢形态、动情周期等变化.测定血清黄体生成素、睾酮、胰岛素、雌激素.免疫组化法分析两组大鼠卵巢中SLPI的表达,RT-PCR及Western印迹技术检测并比较两组大鼠SLPI mRNA及蛋白质的表达.结果 实验组大鼠卵巢形态及激素变化符合PCOS的诊断,免疫组化结果显示实验组较正常对照组大鼠SLPI表达增加,多囊卵巢的卵巢基质、黄体细胞、卵泡的颗粒层及膜中皆有SLPI表达.实验组大鼠卵巢SLPI的基因(P<0.05)及蛋白表达(P<0.05)均高于正常对照组大鼠.结论 多囊卵巢中SLPI的表达增加可能与PCOS大鼠卵巢功能修复有关.

Objective To study the expression and biological effects of secretory leukocyte protease inhibitor (SLPI) in polycystic ovary syndrome (PCOS) of rat model.Methods Sixteen female Sprague-Dawley(SD) rats were divided into two groups:experimental group and normal control group(8 in each group).PCOS were induced in experimental group by combination of insulin and human chorionic gonadotrophin,while rats in control group were injected with physiological saline.The ovary morphological changes and estrous cycle were observed and compared.Serum luteinizing hormone,testosterone,insulin,estrogen levels were measured.The expression of SLPI in the ovary of each group were detected by immunohistochemistry,and SLPI mRNA and protein were measured and compared by RT-PCR and Western blot.Results The changes of ovary morphology and sex hormone levels conformed to clinical PCOS.Immunohistochemical analysis showed that SLPI expression was higher in the ovarian stroma,corpus luteum,and granulose layer of the follicle in polycystic ovary (PCO) compared with that in the normal ovary.The levels of SLPI mRNA and protein were higher in experimental group than those in the normal control group.Conclusion An increase of SLPI expression in PCO may be related to the suppression of ovary disruption in rats with PCOS.
