用核酸原位杂交和用免疫组织化学法检测了6例豚鼠正常肺组织、10~18例癌旁正常肺组织和14例正常人外周血端粒酶逆转录酶(TERT) 亚基的mRNA和相应蛋白质表达.结果显示,豚鼠和人体正常肺组织支气管上皮细胞,肺泡巨噬细胞和少数肺泡上皮细胞有TERT mRNA和蛋白质表达.正常人外周血淋巴细胞中TERT原位杂交阳性表达细胞数为(17.26±8.57)%,免疫组织化学阳性细胞表达数为(28.10±16.78)%.提示:①正常肺组织支气管上皮细胞、肺泡巨噬细胞、少数肺泡上皮细胞和外周血淋巴细胞有TERT mRNA和蛋白质表达;②正常肺组织中上述细胞TERT mRNA和蛋白质的表达在生理状况下对于保证支气管上皮和肺泡上皮细胞再生,维持上皮组织的完整性可能起着重要的作用,同时有利于肺泡巨噬细胞发挥其肺内"卫士”功能;③淋巴细胞TERT表达对于维持机体的免疫功能可能有重要作用.
用覈痠原位雜交和用免疫組織化學法檢測瞭6例豚鼠正常肺組織、10~18例癌徬正常肺組織和14例正常人外週血耑粒酶逆轉錄酶(TERT) 亞基的mRNA和相應蛋白質錶達.結果顯示,豚鼠和人體正常肺組織支氣管上皮細胞,肺泡巨噬細胞和少數肺泡上皮細胞有TERT mRNA和蛋白質錶達.正常人外週血淋巴細胞中TERT原位雜交暘性錶達細胞數為(17.26±8.57)%,免疫組織化學暘性細胞錶達數為(28.10±16.78)%.提示:①正常肺組織支氣管上皮細胞、肺泡巨噬細胞、少數肺泡上皮細胞和外週血淋巴細胞有TERT mRNA和蛋白質錶達;②正常肺組織中上述細胞TERT mRNA和蛋白質的錶達在生理狀況下對于保證支氣管上皮和肺泡上皮細胞再生,維持上皮組織的完整性可能起著重要的作用,同時有利于肺泡巨噬細胞髮揮其肺內"衛士”功能;③淋巴細胞TERT錶達對于維持機體的免疫功能可能有重要作用.

In situ hybridization and immunohistostaining methods were employed to detect the expression of mRNA and protein of telomerase reverse transcriptase in normal lung tissue of 6 guinea pigs, adjacent normal tissues of 10-18 cases of carcinoma and circulating lymphocytesof 14 healthy subjects. The results showed that TERT mRNA and protein expression were detectable in epithelial cells of bronchi, pulmonary alveolar epithelial cells and alveolar macrophages of normal lung tissue in guinea pig and human body. In healthy subjects, the circulating lymphocytes positive for TERT expression were (17.26±8.57) % by in situ hybridization and (28.10±16.78) %, respectively. It was suggested that the expression of TERT mRNA and protein in some actively regenerating normal pulmonary cells might play a critical physiological role in keeping regeneration and integrality of airway epithelial tissue and pulmonary alveolar epithelium. It was also very important for alveolar macrophage to play as "safeguard” function in normal lung. The expression of TERT in circulating lymphocytes might play a critical role in maintaining normal human immunological function.
