目的 研究乳腺癌患者外周血中趋化因子及其受体表达变化在乳腺癌发病机制中的作用.方法 采用酶联免疫吸附试验(ELISA)检测30例乳腺癌患者及健康者血清中单核细胞趋化蛋白1(MCP-1)、活化后可调节的、正常T细胞表达和分泌物因子(RANTES),白介素8(IL-8)及间质细胞衍生因子1(SDF-1)等水平变化.同时用流式细胞仪分析外周血 CD3~+T淋巴细胞表面趋化因子受体CCR2、CCR5、CXCR1、CXCR4的表达.结果 与健康者相比,乳腺癌患者血清中SDF-1及RANTES水平显著升高(均P<0.01),外周血CD3~+T淋巴细胞表面趋化因子受体表达水平无显著性差异.结论 乳腺癌患者血清SDF-1及RANTES水平升高可能在乳腺癌的发病及进展过程中发挥作用.

Objective To study the role of serum chemokines and its receptors in the pathogenesis of breast cancer. Methods Serum levels of monocyte chemotactic protein 1 (MCP-1), regulation on activation normal T-cell expression and secretion(RAN-TES), interleukin 8(IL-8), and the stromal cell derived from factor-1 (SDF-1)were determined by enzyme-linked immunosorbent as-say(ELISA) in 30 patients with breast cancer and 30 normal controls. Meanwhile, chcmokinc receptors, CCR2, CCR5, CXCR1 and CXCR4 on peripheral blood CD3 ~+ -lymphocytes from 20 patients and 20 normal controls were analyzed by flow cytometry with two-color immunofluorescent stains. Results Serum levels of SDF-1 and RANTES were significantly higher in patients than those in controls(P<0. 01). There was no significant difference in expressions of CCR2,CCR5,CXCR1 and CXCR4 on pcripheral blood CD3~+ T-lymphocytes between patients and controls. Conclusion The increase of levels of SDF-1 and RANTES may play an impor-tant role in the occurrence and development of breast cancer.
