目的 探讨孕期炎症刺激对子代大鼠血管局部肾素血管紧张素系统的影响。方法 Sprague Dawley(SD)孕鼠6只随机分为对照组和脂多糖(LPS)组,于孕期第8、10、12天分别给予生理盐水和LPS 0.79 mg/kg(腹腔注射),子代大鼠出生后采用无创伤尾动脉方法测定血压,免疫组织化学方法测定胸主动脉血管紧张素Ⅱ(AngⅡ)蛋白的表达水平,Western blot方法测定血管局部血管紧张素Ⅱ1型受体( AT1 R)、血管紧张素Ⅱ2型受体(AT2R)的蛋白含量。结果 LPS组子代大鼠出生后第12周血压明显高于对照组子代大鼠(P<0.01),15周龄胸主动脉血管组织的AngⅡ表达明显高于对照组子代大鼠(P<0.01),AT1R蛋白表达明显高于对照组子代大鼠(P<0.05),AT2R蛋白表达低于对照组子代大鼠,但差异无统计学意义,AT1R/AT2R比值明显高于对照组子代大鼠(P<0.01)。结论 孕期炎症刺激导致子代大鼠血管局部肾素血管紧张素系统异常,可能是子代大鼠高血压发生发展的重要原因。

목적 탐토잉기염증자격대자대대서혈관국부신소혈관긴장소계통적영향。방법 Sprague Dawley(SD)잉서6지수궤분위대조조화지다당(LPS)조,우잉기제8、10、12천분별급여생리염수화LPS 0.79 mg/kg(복강주사),자대대서출생후채용무창상미동맥방법측정혈압,면역조직화학방법측정흉주동맥혈관긴장소Ⅱ(AngⅡ)단백적표체수평,Western blot방법측정혈관국부혈관긴장소Ⅱ1형수체( AT1 R)、혈관긴장소Ⅱ2형수체(AT2R)적단백함량。결과 LPS조자대대서출생후제12주혈압명현고우대조조자대대서(P<0.01),15주령흉주동맥혈관조직적AngⅡ표체명현고우대조조자대대서(P<0.01),AT1R단백표체명현고우대조조자대대서(P<0.05),AT2R단백표체저우대조조자대대서,단차이무통계학의의,AT1R/AT2R비치명현고우대조조자대대서(P<0.01)。결론 잉기염증자격도치자대대서혈관국부신소혈관긴장소계통이상,가능시자대대서고혈압발생발전적중요원인。
Objective To explore the impact of prenatal exposure to lipopolysaccharide on reninangiotensin system of offspring rats Methods Six pregnant SD rats were randomly divided into 2 groups.The rats in the lipopolysaccharide (LPS) group were treated with LPS 0. 79 mg/kg (I. P. ) on the 8th, 10th and 12th day of gestation, and rats in the control group were treated with saline at the same time points. The blood pressure of offspring rats was measured by the tail cuff method. Protein expression of Angiotensin Ⅱ (Ang Ⅱ ) in thoracic aorta vessel was determined by immunohistochemisry. Protein expressions of Ang Ⅱtype 1 and type 2 receptor in thoracic aorta vessel were detected by Western blot. Results Blood pressure of 12-week-old offspring rats of LPS group was significantly higher than that of 12-week-old offspring rats of control group( P < 0. 01 ). The protein expression of Ang Ⅱ and Ang Ⅱ type 1 receptor in thoracic aorta vessel were significantly higher while protein expression of Ang Ⅱ type 2 receptor was lower in 15-week-old offspring rats of LPS group than in control group, resulting in a significant increase in the ratio of Ang Ⅱtype 1 receptor/Ang Ⅱ type 2 receptor in the aorta at 15-week-old of offspring rats than in 15-week-old offspring rats of control group( P < 0. 01 ). Conclusion Prenatal lipopolysaccharide exposure results vascular renin-angiotensin system dysfunction, which my play an important role on the pathogenesis of hypertension development in offspring rats.
