目的 研究和克隆原发性肝细胞癌(HCC)相关基因,探索其在肝癌发病中的临床意义.方法 采用mRNA差异显示技术,分析肝细胞癌和癌旁正常肝组织间基因的差异表达,获得差异表达基因片段,对其进行克降,测序,并用RT-PCR方法,检测所获得的基因在44对肝细胞癌及相应痛旁正常肝组织中差异表达的情况,分析该基因表达与HCC临床病理特征的关系.结果 克隆了一个肝癌相关基因EEG1,其在肝癌组织中的表达水平明显低于癌旁止常肝组织,仅为正常组织表达量的0.586±0.346倍(P<0.05).EEG1基因在肝癌组织中的表达下调与肝癌的早期分化程度密切相关(P<0.05).结论 差异表达基因EEG1可能在HCC的早期发病中起着重要作用,有望成为肝癌的早期诊断指标.

목적 연구화극륭원발성간세포암(HCC)상관기인,탐색기재간암발병중적림상의의.방법 채용mRNA차이현시기술,분석간세포암화암방정상간조직간기인적차이표체,획득차이표체기인편단,대기진행극강,측서,병용RT-PCR방법,검측소획득적기인재44대간세포암급상응통방정상간조직중차이표체적정황,분석해기인표체여HCC림상병리특정적관계.결과 극륭료일개간암상관기인EEG1,기재간암조직중적표체수평명현저우암방지상간조직,부위정상조직표체량적0.586±0.346배(P<0.05).EEG1기인재간암조직중적표체하조여간암적조기분화정도밀절상관(P<0.05).결론 차이표체기인EEG1가능재HCC적조기발병중기착중요작용,유망성위간암적조기진단지표.
Objective To clone and study primary hepatocellular carcinoma(HCC)-related gene,and explore its clinical significance in the pathogenesis of HCC.Methods mRNA differential display technology was applied to detecting the differential expression in HCC tissue and cancer adjacent normal liver tissue.The differential expressed gene fragment was acquired.Its PCR clones were scquenced and checked for significant alignments in GenBank.The distribution in normal liver tissue and HCC tissue was examined by RT-PCR.The correlation was analyzed between the differential expressed gene and clinicopathologic characleristics of HCC.Results A HCC-related gene EEG1 was cloned.As compared with that in normal liver tissue,EEG1's expression level in HCC tissue was significantly lower(only 0.586 ± 0.346 times,P<0.05).Downregulation of EEG1 in HCC tissue was closely associated with early differentiation of HCC(P<0.05).Conclusion The differential expressed gene EEG1 may play an important role in onset of HCC,which may be used as a potential early tumor gene marker for the diagnosis of HCC.
