整合素(Integrins)是细胞膜蛋白的组成万分,它们是一类细胞粘附分子,并与形态形成的调节密切相关.整合素在细胞与细胞之间,细胞与基质之间的粘附中发挥着重要的作用.本研究通过选择使用鼠抗人整合素β抑止性单克隆抗体和刺激性单克隆抗体,利用噬菌体展示技术,成功地构建了单链Fv抗体(scFv),并对其与整合素β1的结合,选择最佳的可溶性抗体的生产条件等诸方面,进行了应用性研究.同时,也应用链转换(China shuffling)试图改变抗体亲和力进行了探索的试验.

정합소(Integrins)시세포막단백적조성만분,타문시일류세포점부분자,병여형태형성적조절밀절상관.정합소재세포여세포지간,세포여기질지간적점부중발휘착중요적작용.본연구통과선택사용서항인정합소β억지성단극륭항체화자격성단극륭항체,이용서균체전시기술,성공지구건료단련Fv항체(scFv),병대기여정합소β1적결합,선택최가적가용성항체적생산조건등제방면,진행료응용성연구.동시,야응용련전환(China shuffling)시도개변항체친화력진행료탐색적시험.
Integrins belong to one group of adhesion molecules, which are major components in the organization of the cell membrane Integrins are closely associaed with the regulation during morphogenesis and play some important role in biological mechanisms such as signal transduction[1,2]. In this report, the scFv were generated by phage display antibody system.The murine hybridomas, which secrete anti-human β1 stimulative monoclonal antibody( McAb)or inhibitory McAb separately,were used as a source for scFv construction. The investigation was carried out for reactions of engineering antibodies to human β1 integrin and the parameters related to the optimum production of soluble scFv in bacterial culture Meanwhile, the possible improvement of antibody affinity was attempted by using chain shuffling technique.
