目的 观察磷脂酶Cε1(PLCE1)基因过表达对结肠癌细胞生长增殖的影响.方法 通过基因转染、0.7 g/L浓度的G418压力维持筛选出PLCE1过表达100倍以上的SW620结肠癌细胞株;通过噻唑蓝(MTT)生长曲线、克隆形成、裸鼠成瘤等实验观察PLCE1基因过表达对结肠癌细胞生长及增殖的影响.结果 PLCE1基因过表达导致结肠癌细胞生长速度减慢,实验组平板克隆吸光度(A)值为0.3225±0.0065,软琼脂克隆数为11.80 ±2.56,肿瘤质量(0.317±0.169)g,均较亲本组和对照组显著下降,差异有统计学意义(P<0.05),提示转染PLCE1基因后细胞恶性程度降低.结论 PLCE1过表达抑制结肠癌细胞的生长,该基因可能是新的结直肠癌相关抑癌基因.
目的 觀察燐脂酶Cε1(PLCE1)基因過錶達對結腸癌細胞生長增殖的影響.方法 通過基因轉染、0.7 g/L濃度的G418壓力維持篩選齣PLCE1過錶達100倍以上的SW620結腸癌細胞株;通過噻唑藍(MTT)生長麯線、剋隆形成、裸鼠成瘤等實驗觀察PLCE1基因過錶達對結腸癌細胞生長及增殖的影響.結果 PLCE1基因過錶達導緻結腸癌細胞生長速度減慢,實驗組平闆剋隆吸光度(A)值為0.3225±0.0065,軟瓊脂剋隆數為11.80 ±2.56,腫瘤質量(0.317±0.169)g,均較親本組和對照組顯著下降,差異有統計學意義(P<0.05),提示轉染PLCE1基因後細胞噁性程度降低.結論 PLCE1過錶達抑製結腸癌細胞的生長,該基因可能是新的結直腸癌相關抑癌基因.

Objective To observe the effects of phospholipase C epsilon-1 (PLCE1) over-expression on growth and proliferation of colon cancer cells.Methods The stable SW620 colon cancer cells with 100 times of PLCE1 up-regulation were constructed by gene transfection.The effects of PLCE1 over-expression on cancer cell malignant degree were measured by methyl thiazol tetrazolium (MTT) assay,plate colony formation assay,soft agar colony formation assay and xenograft assay.Results Over-expression of PLCE1 significantly inhibited the growth and proliferation of colon cancer cells.In PLCE1 over-expression group,absorbance value of plate colony formations was 0.3225 ± 0.0065,number of soft agar colony formations was 11.80 ±2.56 and the tumor quality was (0.317 ±0.169) g.There was significant difference between experimental group and control group.The cell malignant degree after transfection was reduced.Conclusion These results suggested that PLCE1 may be involved in the development of sporadic colorectal cancer through its inhibitory effect on cell proliferation.PLCE1 exhibited a suppressive role in incidence of colorectal cancer.
