采用Sevag法、木瓜蛋白酶-Sevag法、TCA-Sevag法对绞股蓝粗多糖脱蛋白工艺进行研究.通过DEAE—52纤维素柱层析和葡聚糖凝胶G—200柱层析对粗多糖进行分离纯化,采用高效液相色谱分析法对精制多糖GPM—21的相对分子质量进行测定分析.结果表明:木瓜蛋白酶-Sevag法效果最佳,蛋白脱除率高且多糖损失量小,累计蛋白脱除率可达63.08%,多糖保留率为77.34%.液相渗透色谱法测定GPM—21的相对分子质量为200 576.

The optimum technologies of deproteinization using Sevag method,Papain-Sevag method and TCA-Sevag method were studied.In addition,crude polysaccharides were isolated and purified by DEAE—52 cellulose column chromatography,and Sephadex G—200 column chromatography,and relative molecular mass of refined polysaccharides GPM—21 were measured by HPLC.The results indicated that papain-Sevag method was the best method for the deproteinization in among of methods.The removal rate of total protein could reach up to 63.08%,and the retention rate of polysaccharide was 77.34%.The relative molecular mass of GPM—21 was 200 576.
