设计并制作了可容纳塑料离心管的新型分光光度计液池.液池适用的样品体积变化范围宽(10~500 μL),尤其适合于生物临床样品、强碱性介质以及大批样品的分析.该装置应用于血清蛋白质(总蛋白、白蛋白)、尿素氮、葡萄糖的测定及聚合酶链式反应(PCR)反应管的均一性检验,取得了满意的结果.

A new cuvette which is able to contain a plastic centrifuge tube or a PCR tube for directly spectrophotometric detection was designed and tested. Absorption characteristics of a plastic centrifuge tube in the cuvette was investigated. The tubes showed obvious absorption in ultra-violet region at wavelength less than 300 nm, indicating that the detection wavelength should be set above 400 nm when applying the cuvette for the assay of samples. Plastic tube in this new-designed cuvette showed excellent linear response. Based upon the above investigations, this new-designed cuvette was applied to the determination of total protein, albumin, glucose and urea in human sera, the results were in good agreement with those of clinical assays. Besides, homogeneity of PCR tubes has been examined, and the results suggested that PCR tubes with a same batch number should be used for performing an accurate PCR experiment. In summary, this new-designed cuvette bears the merits of simplicity for use and variable sample volume in a wide range (10~500 μL), and is suitable for samples with large number. Further, contamination can be avoided for biological and clinical samples by using this cuvette.
