目的 探讨秋水仙碱对压力负荷性大鼠心肌中转化生长因子β_1(TGF-β_1)的影响.方法 利用腹主动脉缩窄的方法 建立压力超负荷性心肌大鼠模型,放射免疫分析法测定局部心肌组织中血管紧张素Ⅱ(Ang Ⅱ)水平,RT-PCR法半定量TGF-β_1mRNA的表达变化,免疫组织化学法测定心肌TGF-β_1的表达,同时给予秋水仙碱8周,观察上述指标的变化.结果 与对照组相比,模型组AngⅡ、TGF-β_1 mRNA和TGF-β_1 表达明显增加(P<0.01),秋水仙碱能明显减少上述指标的表达.结论 秋水仙碱可以通过抑制压力负荷性心室重构大鼠RAAS和交感神经系统活性,减少神经内分泌因子AngⅡ生成,减少纤维化因子TGF-β_1的表达.

목적 탐토추수선감대압력부하성대서심기중전화생장인자β_1(TGF-β_1)적영향.방법 이용복주동맥축착적방법 건립압력초부하성심기대서모형,방사면역분석법측정국부심기조직중혈관긴장소Ⅱ(Ang Ⅱ)수평,RT-PCR법반정량TGF-β_1mRNA적표체변화,면역조직화학법측정심기TGF-β_1적표체,동시급여추수선감8주,관찰상술지표적변화.결과 여대조조상비,모형조AngⅡ、TGF-β_1 mRNA화TGF-β_1 표체명현증가(P<0.01),추수선감능명현감소상술지표적표체.결론 추수선감가이통과억제압력부하성심실중구대서RAAS화교감신경계통활성,감소신경내분비인자AngⅡ생성,감소섬유화인자TGF-β_1적표체.
Objective To investigate the effect of colchicine on myocardium β_1.Methods The model was induced by abdominal aorta constriction(AC)in rats.After 8 weeks treatment,cardiac muscle AngⅡwere measured by radioimmunoassay,and ventricular transforming growth factor-beta 1(TGF-β_1)was examined bv RT-PCR,and the content of TGF-β_1 in myocardium were determined using immunohistochemical analysis after AC.Results The levels of Ang Ⅱ and TGF-β_1 mRNA and the content of TGF-β_1 were significantly increased after AC eight weeks and significantly decreased after treatment with colchicine.Conclusion Colchicine can significantly attenuate the level of myocardium TGF-β_1;the mechanism is related with its ability to inhibit the activation of rennin-angiontensin-aldosterone system (RAAS)and systema nervosum sympatheticum(SNS),decrease the content of Ang Ⅱ and TGF-β_1.
