Plasmacytoid dendritic cells (pDC) produce high levels of interferon (IFN)-α in response to viral infection in humans and act as an important link between innate and adaptive immunity. IFN-α production in patients infected with HIV-1 is deficient. The mechanisms for the decreased circulating pDC and their dysfunction in HIV-infected patients are unknown. Whether IFN-α is beneficial or detrimental in HIV infection remains a matter of debate. The balance between pDC numbers and function is important in the pathogenesis of HIV-1 infection. Therefore, using absolute pDC counts and function as a biomarker, along with CD4+ cell counts and viral load in HIV-1-infected patients has potential value.
