目的 观察颐和春口服液对肾阳虚大鼠睾丸功能的影响并探讨其机制.方法 用腺嘌呤制备肾阳虚大鼠模型.实验分为正常对照组、肾阳虚模型组、阳性药物治疗组和颐和春治疗组.观察颐和春口服液对肾阳虚大鼠睾丸形态学、血清雄性激素及睾丸组织MDA和SOD的影响,并观察对腺垂体细胞凋亡及下丘脑区兴奋性及抑制性氨基酸比值的影响.HE染色观察睾丸形态学改变、放免法测定血清雄性激素、生化法测定睾丸组织MDA和SOD,末端标记法测定腺垂体细胞凋亡,HPLC测定脑组织氨基酸.结果 颐和春明显改善肾阳虚引起的睾丸损伤,提高雄性激素水平,降低睾丸组织MDA,提高SOD,减少腺垂体细胞凋亡,调节下丘脑兴奋性和抑制性氨基酸比值.结论 颐和春口服液对肾阳虚大鼠生殖能力有一定改善作用,可能与其对下丘脑-腺垂体-睾丸轴的调节作用有关.

Objective To observe the effect of Yihechun oral liquid (YOL)on testicular function of kidney-yang deficiency rats and to explore its mechanism. Methods The kidney-yang deficiency rat model was induced by gastric infusion of adenine. The rats were divided into four groups: normal control group, kidney-yang deficiency model group, positive drug group, and YOL group. We observed the effect of YOL on testis morphology, serum androgen level, MDA and SOD activities in testicular tissue. Meanwhile, the apoptosis of adenopituicytes and the ratio of excitatory and inhibitory amino acids in brain were also observed. Testicular morphological changes were observed by HE staining, radioimmunoassay was used to detect serum androgen, and biochemical determination was used to observe MDA and SOD activities in testicular tissue. The adenopituicyte cell apoptosis was detected by terminal-labeling method, amino acid content in brain tissue was determined by HPLC. Results YOL obviously relieved testicular damage caused by kidney-yang deficiency, increased the testosterone levels, decreased the MDA, increased the SOD, decreased the adenopituicyte cell apoptosis , and regulated the ratio of excitatory and inhibitory amino acids in the hypothalamus. Conclusion YOL has certain effect on improving reproductive capacity of kidney-yang deficiency rats, which is probably related to the regulation of hypothalamus-pituitary - testicular axis.
