目的 观察β-七叶皂苷钠在减轻大鼠高压电损伤后炎症反应中的作用.方法 将90只SD大鼠随机分为3组,分别于伤前30min静脉注射β-七叶皂苷钠(β-七叶皂苷钠组,AC组)、地塞米松(地塞米松组,DX组)及生理盐水(生理盐水组,SC组),均制成下肢高压电损伤模型.每组于伤后1、6、24h各开胸处死10只大鼠,抽血检测中性粒细胞比值,酶联免疫吸附法测肿瘤坏死因子a(TNF-α)及白细胞介素6(IL-6)含量,取创周组织行组织学观察.结果 伤后1h各组IL-66、TNF-α比较,差异无统计学意义(P>0.05).伤后6hAC组和DX组IL-6分别为(138.66±21.56)pg/mL、(142.43±25.63)pg/mL,TNF-α分别为(160.86±24.22)pg/mL、(128.76±18.63)pg/mL,均明显低于SC组的(217.63±40.24)pg/mL和(266.75±45.62)pg/mL,差异有高度统计学意义(P<0.01).AC 组和DX组间IL-6、TNF-α水平差异无统计学意义(P>0.05).伤后24h AC组和DX组IL-6和TNF-α低于SC组,且AC组较DX组表达下调.各时相点AC组及DX组血中性粒细胞比值均低于SC 组(P<0.05或P<0.01).结论 β-七叶皂苷钠干预可减轻大鼠高压电损伤后的炎症反应.

목적 관찰β-칠협조감납재감경대서고압전손상후염증반응중적작용.방법 장90지SD대서수궤분위3조,분별우상전30min정맥주사β-칠협조감납(β-칠협조감납조,AC조)、지새미송(지새미송조,DX조)급생리염수(생리염수조,SC조),균제성하지고압전손상모형.매조우상후1、6、24h각개흉처사10지대서,추혈검측중성립세포비치,매련면역흡부법측종류배사인자a(TNF-α)급백세포개소6(IL-6)함량,취창주조직행조직학관찰.결과 상후1h각조IL-66、TNF-α비교,차이무통계학의의(P>0.05).상후6hAC조화DX조IL-6분별위(138.66±21.56)pg/mL、(142.43±25.63)pg/mL,TNF-α분별위(160.86±24.22)pg/mL、(128.76±18.63)pg/mL,균명현저우SC조적(217.63±40.24)pg/mL화(266.75±45.62)pg/mL,차이유고도통계학의의(P<0.01).AC 조화DX조간IL-6、TNF-α수평차이무통계학의의(P>0.05).상후24h AC조화DX조IL-6화TNF-α저우SC조,차AC조교DX조표체하조.각시상점AC조급DX조혈중성립세포비치균저우SC 조(P<0.05혹P<0.01).결론 β-칠협조감납간예가감경대서고압전손상후적염증반응.
Objective To investigate the effect of β-aescin on the inflammatory reaction after electrical injury in rats.Methods Experimental animal models of electrical injury were made in 90 rats.The rats were randomly divided into 3 groups as the followings:β-aescin group (AC),dexamethasone group (DX) and normal saline group (SC),which were respectively given β-aescin,dexamethasone and normal saline by caudal vein 30 minutes before the injury.Ten rats of each groups were sacrificed at 1 hour,6 hours and 24 hours after the injury.Blood samples were collected immediately to measure tumor necrosis factor-alpha (TNF-α),interleukin-6 (IL-6) and neutiophil percentage.Tissue samples around the wounds were harvested,HE stained and observed by microscope.Results There was no significant difference among the three groups for mean values of IL-6 and TNF-α at 1 hour.Mean values of IL-6 and TNF-α at 6hours of AC group and DX group were respectively (138.66 ± 21.56) pg/mL,(160.86 ± 24.22) pg/mL and (142.43 ± 25.63 ) pg/mL,(128.76 ± 18.63 ) pg/mL,with that of AC and DX group much lower than SC group[ (217.63 ±40.24)pg/mL and (266.75 ±45.62) pg/mL] (P<0.01).Mean values of IL-6 and TNF-α at 24 hours of AC group and DX group were both lower compared with that of SC group and there were significant differences between the AC group and DX group for the value of IL-6 and TNF-α.The mean neutrophil percentages of AC group and DX group at all the time points were much lower than that of the SC group ( P < 0.05 for 1 h and 6 h,P < 0.01 for 24 h).There was no significant difference between AC group and DX group for the three items.Conclusions The inflammatory reaction of electrically injured rats can be depressed by β-aescin interference.
