The major limitation to a long-term use of opioids, such as morphine, is the development of physiological tolerance,ob-served as a profound decrease in analgesic effect. Although the molecular and cellular mechanisms underlying the development of mor-phine tolerance remain controversial, great progress has been made. This review focuses on the opioid receptor with the aim of gaining a greater insight into the mechanism of morphine tolerance to analgesia.
