目的 了解广西两地理株钉螺细胞色素氧化酶Ⅰ(cytochrome oxidase Ⅰ,CO Ⅰ)基因的差异性,探讨广西钉螺与周边邻近4省钉螺的亲缘关系.方法 采集广西靖西有肋、横县光壳钉螺,提取DNA,PCR扩增CO Ⅰ基因并测序.登录GenBank检索出云南大理、四川绵竹、湖南岳阳、湖北汉阳4地钉螺的CO Ⅰ基因序列.采用Clustal X软件对上述CO Ⅰ基因排序.采用拟钉螺CO Ⅰ基因序列作为外源性参考基因,Kimura双参数法计算遗传距离,Mega3.1软件中非加权组平均法(UPGMA)、邻接法(NJ)和最小进化法(ME)构建系统发生树.结果 广西靖西光壳钉螺与横县有肋钉螺CO Ⅰ基因的遗传距离为0.0505,同源性为95%.广西横县钉螺与湖南岳阳、湖北汉阳、四川绵竹、云南大理钉螺的遗传距离依次为0.0102、0.0171、0.1401、0.1421,CO Ⅰ基因的同源性依次为99%、98%、87%、86%.广西靖西钉螺与湖南岳阳、湖北汉阳、四川绵竹、云南大理钉螺的遗传距离依次为0.0451、0.0505、0.1382、0.1421,C0 Ⅰ基因的同源性依次为95%、95%、88%、87%.结论 广西有肋钉螺与光壳钉螺基因差异性较小.广西两种钉螺与湖南、湖北钉螺遗传距离较小,同源性较高.广西两种钉螺与四川、云南钉螺遗传距离较大,同源性较低.

Objective To investigate the genetic difference of cytochrome oxidase Ⅰ (CO Ⅰ ) of two isolates of Oncomelania hupensis collected from Guangxi, and analyze the phylogenic relationship with isolates from 4 neighboring provinces. Methods Ribbed-shelled and smooth-shelled snails were collected from Jingxi County and Hengxian County of Guangxi Zhuang Autonomous Region respectively. Genomic DNA was extracted from the snails. CO Ⅰ gene fragments were amplified by PCR,then purified and sequenced. The data of CO Ⅰgene sequences of Yunnan,Sichuan,Hunan,and Hubei isolates were obtained from the GenBank. Sequence of each isolate was edited by using Clustal X software. The CO Ⅰ gene of Tricia was used as the reference of exogenous gene. The genetic distance was computed with Kimura method. The phylogenetic tree was constructed by the unweighted pair group method with arithmetic mean (UPGMA) as well as the methods of neighbor-joining (NJ) and minimum evolution (ME) with the MEGA (3.1) software. Results The genetic distance and homology of CO Ⅰ between Jingxi isolates (ribbed-shelled) and Hengxian isolates (smooth-shelled) was 0. 0505 and 95% ,respectively. The genetic distances of Hengxian isolate with other isolates from Yunnan, Sichuan, Hunan ,and Hubei Province were 0. 0102,0. 0171,0. 1401 and 0. 1421 ,with the homology of 99% ,98% ,87% and 86% respectively. Correspondingly,the genetic distances of Jingxi isolate with isolates from Yunnan,Sichuan,Hunan,and Hubei province were 0. 0451,0. 0505,0. 1382,and 0. 1421 ,while the homology was 95% ,95%,88% and 87% respectively. Conclusion No significant genetic difference was found between the ribbedshelled and smooth-shelled Oncomelania hupensis isolates from Guangxi Zhuang Autonomous Region. They were phylogenetically closer,and more homologous to the isolates from Hunan and Hubei Province. However,they showed longer genetic distance and less homologous to the isolates from Sichuan and Yunnan Province.
