目的选择脊髓灰质炎病毒繁殖的最佳温度,同时观察温度对病毒繁殖及D抗原的影响.方法分别在25℃、33℃和37℃培养脊髓灰质炎病毒(Sabin株Ⅰ、Ⅱ和Ⅲ型),从24 h开始间隔12 h取样,至细胞完全病变或96 h,用ELISA检测病毒D抗原含量,绘制变化曲线;用微量细胞病变法检测感染性滴度,绘制病毒生长曲线.结果三个型的脊髓灰质炎病毒在25℃均不繁殖;33℃和37℃均适合病毒繁殖.对于Ⅰ和Ⅲ型病毒,33℃的繁殖滴度及D抗原水平比37℃高;Ⅱ型病毒在两种温度下繁殖特性基本一致.结论温度对脊髓灰质炎病毒繁殖起决定性作用,综合Ⅰ、Ⅱ和Ⅲ型病毒的温度繁殖特点,33℃培养比37℃更适合脊髓灰质炎病毒繁殖.

Objective To determine the optimal temperature for poliovirus propagation and D-anti-gen production. Methods Poliovirus(Sabin strain)type Ⅰ、Ⅱ and Ⅲ were cultivated on Veto cells at 25℃ ,33℃ ,and 37℃,respectively,and sampled at an interval of 12 h until the cells aggregated com-pletely or 96 h after inoculation of the cells. D-antigen was assessed with ELISA,poliovirus were titrated with the micro-cell plaque assays. Both diagrams were presented for either cultivation or D-antigen pro-duction analysis. Results No virus propagation were detected in the cell supernatant at 25 ℃ for all the three types of poliovirus,however,virus propagation were prominent at both 33℃ and 37 ℃,while 33 ℃ being optimal for type Ⅰ and Ⅲ in terms of D-antigen production and poliovirus propagation. Type Ⅱ showed no difference between 33 ℃ and 37 ℃ on propagation. Conclusions Temperature is very im-portant for poliovirus propagation and D-antigen production and 33 ℃ is superior to 37 ℃ for poliovirus propagation.
