目的 观察褐藻多糖硫酸酯(FPS)对β-淀粉样肽(1-40)(Aβ_(1-40))右侧脑室注射诱导的阿尔茨海默病(Alzheimet's disease,AD)小鼠认知障碍的作用及机制.方法 小鼠右侧脑室注射凝聚态Aβ_(1-40)3 ng,制作AD模型.实验分为FPS低、中、高剂量治疗组(50,100,200 mg·kg~(-1)·d~(-1))、阳性药盐酸多奈哌齐组(2.5 mg·kg~(-1)·d~(-1))、模型组和假手术组,于造模后24 h分别灌胃给药16 d.术后第11天开始进行避暗试验和水迷宫试验.分别检测小鼠的学习和记忆能力,试验结束后测定皮层、海马组织胆碱乙酰转移酶(ChAT)和乙酰胆碱酯酶(AchE)活力变化.结果 与假手术组相比,模型组小鼠的学习、记忆能力显著下降,脑内ChAT活力和AchE活力显著变化;与模型组对比,低、中、高剂量FPS显著改善AD小鼠空间学习记忆能力,增加皮层和海马内ChAT活力,抑制AchE活力.结论 FPS有改善Aβ_(1-40)诱导的AD模型小鼠空间学习记忆障碍的作用,其机制与升高脑内乙酰胆碱含量有关.

목적 관찰갈조다당류산지(FPS)대β-정분양태(1-40)(Aβ_(1-40))우측뇌실주사유도적아이자해묵병(Alzheimet's disease,AD)소서인지장애적작용급궤제.방법 소서우측뇌실주사응취태Aβ_(1-40)3 ng,제작AD모형.실험분위FPS저、중、고제량치료조(50,100,200 mg·kg~(-1)·d~(-1))、양성약염산다내고제조(2.5 mg·kg~(-1)·d~(-1))、모형조화가수술조,우조모후24 h분별관위급약16 d.술후제11천개시진행피암시험화수미궁시험.분별검측소서적학습화기억능력,시험결속후측정피층、해마조직담감을선전이매(ChAT)화을선담감지매(AchE)활력변화.결과 여가수술조상비,모형조소서적학습、기억능력현저하강,뇌내ChAT활력화AchE활력현저변화;여모형조대비,저、중、고제량FPS현저개선AD소서공간학습기억능력,증가피층화해마내ChAT활력,억제AchE활력.결론 FPS유개선Aβ_(1-40)유도적AD모형소서공간학습기억장애적작용,기궤제여승고뇌내을선담감함량유관.
Objective To study the effect of fucoidan on the learning and memory impairment in Alzheimer's disease (AD)model mice induced by aggregated β-amyloid peptide (1-40)(Aβ_(1-40)). Methods AD mice model was established by injected Aβ1-40 into the right mice ventricle. The model mice were administered with fucoidan (50, 100, 200 mg·kg~(-1)·d~(-1), ig), Donepezil (2.5 mg·kg~(-1)·d~(-1), ig) and daily ig injections of normal saline for 16 days respectively 24 hours after aggregated Aβ_(1-40) intracerebroventricular injection. The cognitive ability of AD model was detected by water amaze test and passive avoidance test from the 11th day. Then the animals were sacrificed and the activity of acetylcholinesterase(AChE)and choline acetyltransferase(CHAT)in the cortical and hippocampal tissue were examined by colorimetric method. Results The ability of learning and memory was significant lowered, and the activities of cerebral CHAT and AChE changed in AD group as compared with the sham-operation group. Fucoidan (50, 100, 200 mg · kg~(-1) · d~(-1), ig) significantly ameliorated the learning and memory impairment induced by Aβ_(1-40) Biochemical analysis showed that fucoidan increased CHAT activity and decreased AChE activity in the cortical and hippocampal tissue. Conclusion Fucoidan can significantly improve the learning and memory impairment in mice with AD induced by A β_(1-40), and the mechanism may be related with the increase of cerebral acetylcholine content.
