目的 观察生长激素对着床期SD大鼠子宫内膜整合素αvβ3(Itgαvβ3)及骨桥蛋白(OPN)表达的影响,以探讨改善子宫内膜容受性的方法.方法 随机将50只雌性SD大鼠平均分成两组,对照组为生理盐水(NS)组,实验组为生长激素(GH)组.取发现阴栓后第4天子宫内膜,用免疫组织化学技术检测子宫内膜Itgαvβ3及OPN的表达,同时首次通过SYBR Green嵌合荧光实时定量PCR技术检测子宫内膜上述两种因子mRNA的表达.结果 GH组子宫内膜Itgαvβ3及OPN的表达呈强阳性,对照组表达弱于GH组,两组差异有统计学意义(Itgαvβ3:77.33±4.23比92.56±16.80,0PN:71.75±8.77比80.73± 13.00,均P<0.05).实时荧光定量PCR结果与免疫组织化学结果相符.结论 生长激素可以增强着床期子宫内膜Itgαvβ3及OPN的表达,可能可改善子宫内膜容受性,提高临床妊娠率.

Objective To investigate the impact of growth hormoue (GH) on the expressions of integrin αvβ3 ( Itgαvβ3 ) and osteopontin ( OPN ) in endometrial tissues in SD rats during the impantation period,and to explore the method to improve the endometrial receptivity.Methods Fifty female SD rats were randomized into two groups,including control group(normal saline,NS group) and experimental group (GH group).The endometrium was extracted at 4 d after the presence of vaginal plug.Thereafter,the pro tein expressions of Itg α[β 3 and OPN were detected by immunohistochemical technique,and the mRNA expressions of the two factors mentioned above firstly by SYBR Green real-time PCR technique.Results The expressions of Itgαv3 and OPN were strongly positive in GH group,which were much weaker in NS group(tgoαv[33:77.33±4.23 vs 92.56± 16.80,OPN:71.75±8.77 vs 80.73± 13.00,all P<0.05).The result of real-time PCR was consistent with immunohistochemical technique.Conclusion GH can enhance the expressions of Itgαvβ3 and OPN in endometrium of implantation period,which may improve endometrial receptivity and increase the pregnancy rate in clinic.
