23日龄非免疫健康罗曼蛋公鸡640羽随机均分为6个不同剂量用药组、1个疫苗对照和1个空白对照组,另取60羽设1个疫苗对照组和1个空白对照组,30羽/组,除空白对照组外均用新城疫Ⅳ系苗滴鼻、点眼免疫(3羽份/羽),37日龄二免。在免疫的同时,6个用药组分别饮水添加6种浓度的的归芪饮,0.5mL/羽,每天1次,连续3d;给药后每天观察各组鸡群精神、活动、采食、饮水和发病死亡情况;分别于免疫后第7、14、21、28、35和42天翼静脉采血,检测血清NDV-HI抗体效价;于首次免疫后第8周选取0.5g/mL、1g/mL、1.5g/mL、2g/mL4个剂量组(50羽/组),1个疫苗攻毒对照组(50羽/组),1个空白攻毒对照组(50羽/组),1个疫苗对照组(30羽/组)和1个空白对照组(30羽/组)进行攻毒保护试验。同时,进行了体外刺激外周血T淋巴细胞转化增殖试验。结果显示,各组给药后未见任何不良反应,各剂量组在各时间点的抗体效价多明显或显著高于疫苗对照组和空白对照组,1g/mL剂量组鸡只末重显著增加,1g/mL、1.5g/mL剂量组日增重显著高于对照组,料重比显著低于对照组,2g/mL剂量组料重比显著低于对照组;1.5g/mL组的攻毒保护率最高,其次为1g/mL剂量组;体外试验结果也显示,归芪饮在合适的剂量可以刺激外周血T淋巴细胞增殖。表明归芪饮临床应用安全,增强细胞免疫和体液免疫的效果确实,可以将1-2g/mL剂量作为临床试验应用剂量,1g/mL剂量最佳。

We chosed 640 healthy Roman cock, which were non-immune and 23 days old, dividing the cock into eight groups which separately were one control group, one immune control group and six drug groups which gave different dose drugs of Gui Qiyin. At the same time we also took another 60 Roman cock, dividing equally into two groups which were one control group and one immune control group. Except the two control groups, all the groups were immuned by Newcastle disease vaccine Ⅳ through nose and eyes using 3 times amount each cock. When the cocks were 37 days old, we immuned them again. At the first immune, gave the six drug groups water to drink , every group's water contained different concentration prescription 5. Every chick had 0.5 mL water containing drug every day. And drinking the chicken continued 3 days. After giving the drugs, watch the spirit, activity, eating food, drinking water and death of the cocks every day. At the 7days, 14 days, 21 days, 28 days, 35 days, 42 days after immune, we collected the blood from under the wing to test the NDV-HI antibody titer of the serum. At the eighth week after the first immune, we used the NDV allantoic fluid to carry on artificial virus attack experiment. We took the groups (50 cock in each group) from above which concentration of using the Gui Qiyin drug separately were 0.5 g/mL, 1.0 g/mL, 1.5 g/mL and 2.0 g/mL. The four groups used the NDV allantoic fluid. Taking the immune control group and control group (50 cocks in each group) also above used the NDV allantoic fluid. We also had one immune control group and one control group taken above(30 cocks in each group). At the same time stimulate the T lymphocytes of Peripheral blood to transform and increase in vitro. The results showed that each group had no adverse reactions. The different groups' antibody titer which used the Gui Qiyin drug was obviously higher than the immune control group and control group at any point. In the 1 g/mL group, the weight of cocks at the end increased greatly. In the 1g/mL、1.5g/mL group, the day's increase of cock was clearly more than the control group, and feed conversion ratio was lower than the control group. In the 2 g/mL group, the feed conversion ratio was lower than the control group. In the 1.5g/mL group, protection rate of virus attack is the highest, then was the group of 1g/mL. The result in vitro showed that prescription 5 in proper concentration could stimulate the T lymphocytes to increase. So we can say that the prescription 5 is safe in the clinical and can increase the effect of cell mediated immunity and humoral immune. So the concentration of Gui Qiyin used in clinical was 1~2mg/mL. The best concentration is 1g/mL.
