The progress of analysis of protein by the use of the self-dimerizatron system of acridine red by the dye combination method is reviewed briefly. As a novel development, the determination of protein by the use of the self-dimerization system of acridine red and the optimum condition for the determination are described in this paper. There is a self-polymerization equilibrium system between the cation of the fluorescent dye-acridine red and its dimer. In the presence of a cationic surfactant, e.g. SDS, the above mentioned equilibrium can be adjusted by the quantitative addition of protein, thus leading to a regular change in fluorescence intensity of the system. Based on this phenomenon, the fluorescent determination of protein is made possible. The significant features of this method are its high sensitivity and rapidity of reaction, wide range of linearity, high accuracy and selectivity.
