High resolution micrographs of cytoplasmic polyhedrosis virus (CPV) embedded in vitreous ice at liquid helium temperature were recorded in a 300 kV electron cryomicroscope (cryoEM) equipped with a field emission gun (FEG). Power spectrum analysis of these micrographs indicates that an image resolution better than 0 6 nm was obtained. We present our three dimensional (3D) reconstruction effort using a recently designed Image Management and Icosahedral ReconstructionSystem (IMIRS), which is the first software package that can carry out cryoEM 3D reconstruction on Microsoft Windows platform. A preliminary 3D reconstructionof the full CPV clearly reveals that the T=1 capsid shell of a radius of 28 5 nm encloses regularly spaced dsRNA layers and is decorated with twelve pentamericturrets extending to a radius of 36 0 nm on its icosahedral vertices.
