目的 研究Tesmin基因的表达特点值.方法 将4d、9d、18d、35d、54d和6月龄小鼠睾丸组织提取RNA,通过RT-PCR分析差异表达基因Tesmin在小鼠睾丸不同发育阶段中的表达.结果 基因芯片结果显示4d、9d、18d、35d、54d和6月龄小鼠睾丸中分别是31.4(A)、34(A),1292(P)、865.6(P)、977.2(P) 和830.7(P),代表Tesmin基因在4d,9d小鼠中无表达,18d龄后开始高表达,RT-PCR结果表明小鼠Tesmin基因在小鼠9d龄及之前的睾丸中没有表达,在18d龄后睾丸中开始高表达,与基因芯片分析结果相一致.结论 Tesmin基因为小鼠年龄依赖性表达基因,小鼠Tesmin基因的表达与小鼠精子发生有很强的一致性,推测该基因可能在精子发生中起重要作用.

Objeetive To investigate the expression of Tesmin in the development of mouse testicle.Methods Testes samples from Balb/c mice at different postnatal days (days 4,9,18,35,54,and 6 months) were collected.Differential genes in the development of mouse testicle were screened by mouse whole genome 430 2.0 Array (Affymetrix Inc.)chip.The interested gene Tesmin was further confirmed by RT-PCR.Results The expression of Tesmin was gradually upregulated following with developmental of mouse testis.The hybridization signal intensities of the tested testis on d 4,d 9,d 18,d 35,d54,and 6 months of postnatal were 31.4(Absent expression,A),34(A),1292 (Present expression,P),856.6(P),977.2(P) and 830.7(P) respectively.The expression of Tesmin was detected on d 18,d 35,d 54,and 6 months,whereas no expression was detected on d 4 and d 9.Same expression pattern of Tesmin was confirmed by RT-PCR analysis.Conclusion Tesmin was a time-dependent expression gene in the development of mouse testis,and its expression trend was identical with postnatal mouse spermatogenesis,which sugggested that Tesmin might be involved in mouse spermatogenesis.
