目的 建立大观霉素直接竞争酶联免疫吸附(ELISA)分析方法.方法 通过大观霉素与载体蛋白偶联,制备免疫原和检测抗原,用杂交瘤技术制备单克隆抗体(mAb),鉴定mAb的特异性,并用于竞争ELISA检测.结果 获得3株能稳定分泌抗大观霉素的单克隆抗体杂交瘤细胞株,大观霉素竞争ELISA检测的下限为1 ng/mL.结论 抗大观霉素的mAb具有抗原结合特异性.可用于大观霉素竞争ELISA免疫学检测.
目的 建立大觀黴素直接競爭酶聯免疫吸附(ELISA)分析方法.方法 通過大觀黴素與載體蛋白偶聯,製備免疫原和檢測抗原,用雜交瘤技術製備單剋隆抗體(mAb),鑒定mAb的特異性,併用于競爭ELISA檢測.結果 穫得3株能穩定分泌抗大觀黴素的單剋隆抗體雜交瘤細胞株,大觀黴素競爭ELISA檢測的下限為1 ng/mL.結論 抗大觀黴素的mAb具有抗原結閤特異性.可用于大觀黴素競爭ELISA免疫學檢測.
목적 건립대관매소직접경쟁매련면역흡부(ELISA)분석방법.방법 통과대관매소여재체단백우련,제비면역원화검측항원,용잡교류기술제비단극륭항체(mAb),감정mAb적특이성,병용우경쟁ELISA검측.결과 획득3주능은정분비항대관매소적단극륭항체잡교류세포주,대관매소경쟁ELISA검측적하한위1 ng/mL.결론 항대관매소적mAb구유항원결합특이성.가용우대관매소경쟁ELISA면역학검측.
Objective To establish an enzyme-linked immunosorbent assay (ELISA) for specti- nomycin detection. Methods Immunogen and antigen were prepared by the coupling of specti- nomycin and carrier protein,and monoclonal antibody (mAb) was prepared by hybridoma tech-nique. The specificity of the mAb was appraised and detected by competing ELISA. Results 3 hybridoma cell strains capable of secreting anti-spectinomycin mAb stably were obtained. The lowest limit of ELISA for spectinomycin detection was 1 ng/mL. Conclusion Anti-spectinomycin mAb has the antigen binding specificity and can be used in competing ELISA for spectinomycin detection.
