目的 观察ABCC4(ATP-binding cassette,sub-family C member4)基因对胃癌细胞系SGC-7901增殖及凋亡的作用.方法 在胃癌细胞系SGC-7901中导入携带ABCC4的RNAi慢病毒载体,应用CELLOMICS检测细胞生长及克隆形成情况,采用流式细胞术检测细胞周期及细胞凋亡情况.结果 SGC-7901细胞经ABCC4基因RNAi有效靶点慢病毒(ABCC4 siRNA)感染后,SGC~7901细胞生长减慢,细胞周期发生改变,处于G1期的SGC7901细胞显著减少,(t=2.752,P<0.05),处于G2/M期的SGC-7901细胞显著增加(t=2.973,P<0.05),同时,细胞凋亡显著增加(t=3.068,P<0.05),细胞克隆形成能力减弱(t=3.534,P<0.05).结论 抑制ABCC4基因可以减缓SGC-7901细胞恶性增殖,促进凋亡.

목적 관찰ABCC4(ATP-binding cassette,sub-family C member4)기인대위암세포계SGC-7901증식급조망적작용.방법 재위암세포계SGC-7901중도입휴대ABCC4적RNAi만병독재체,응용CELLOMICS검측세포생장급극륭형성정황,채용류식세포술검측세포주기급세포조망정황.결과 SGC-7901세포경ABCC4기인RNAi유효파점만병독(ABCC4 siRNA)감염후,SGC~7901세포생장감만,세포주기발생개변,처우G1기적SGC7901세포현저감소,(t=2.752,P<0.05),처우G2/M기적SGC-7901세포현저증가(t=2.973,P<0.05),동시,세포조망현저증가(t=3.068,P<0.05),세포극륭형성능력감약(t=3.534,P<0.05).결론 억제ABCC4기인가이감완SGC-7901세포악성증식,촉진조망.
Objective To observe the effect of ABCC4(ATP-binding cassette, sub-family C member 4) on gastric cancer cell line SGC-7901's proliferation and apoptosis. Methods SGC-7901 cell line were infected by the lentivirus carried the RNAi of ABCC4. were detected the proliferation and clone formation through CELLOMICS. Flow cytometry was applied to detect the cell cycle and apoptosis. Results The infection of SC C-7901 cell line with lentivirus carried the RNAi of ABCC4 alleviated the proliferation of cell line, changed the cell cycle, significantly decreased the cell number of Go/G1 (t=2.752,P<0.05), significantly increased the cell number of G2/M (t=2.973, P<0.05), at the same time significantly increased apoptosis (t= 3.068, P<0.05), and inhibited the formation of cell clone (t = 3.534, P<0.05). Conclusion The inhibition of gene ABCC4 can alleviate the malignant proliferation of SGC-7901 and produce its apoptosis.
