目的 观察辐射及多药耐药基因逆转剂反义寡核苷酸( mdr1ASON)联合5-氟尿嘧啶磁性白蛋白微球(5-Fu-MAMS)靶向治疗耐药裸鼠人胰腺癌模型的效果.方法 构建人胰腺癌耐药细胞株(SW1990/Fu),建立裸鼠人胰腺癌耐药模型,选择适当的磁场,施加于肿瘤表面,瘤内注射mdr1ASON及5-Fu-MAMS,观察肿瘤生长,检测其对耐药裸鼠人胰腺癌的治疗效果.结果 在外加磁场的磁导向作用下,5-Fu-MAMS能定位于肿瘤组织,联合mdr1ASON能显著抑制肿瘤生长,肿瘤体积抑制率达到85.00%,肿瘤重量抑制率达到87.74%,与其他组比较差异有统计学意义(P<0.01).结论 辐射促转染的mdr1ASON联合磁性载药微球对肿瘤细胞具有较好的耐药逆转作用.
目的 觀察輻射及多藥耐藥基因逆轉劑反義寡覈苷痠( mdr1ASON)聯閤5-氟尿嘧啶磁性白蛋白微毬(5-Fu-MAMS)靶嚮治療耐藥裸鼠人胰腺癌模型的效果.方法 構建人胰腺癌耐藥細胞株(SW1990/Fu),建立裸鼠人胰腺癌耐藥模型,選擇適噹的磁場,施加于腫瘤錶麵,瘤內註射mdr1ASON及5-Fu-MAMS,觀察腫瘤生長,檢測其對耐藥裸鼠人胰腺癌的治療效果.結果 在外加磁場的磁導嚮作用下,5-Fu-MAMS能定位于腫瘤組織,聯閤mdr1ASON能顯著抑製腫瘤生長,腫瘤體積抑製率達到85.00%,腫瘤重量抑製率達到87.74%,與其他組比較差異有統計學意義(P<0.01).結論 輻射促轉染的mdr1ASON聯閤磁性載藥微毬對腫瘤細胞具有較好的耐藥逆轉作用.
목적 관찰복사급다약내약기인역전제반의과핵감산( mdr1ASON)연합5-불뇨밀정자성백단백미구(5-Fu-MAMS)파향치료내약라서인이선암모형적효과.방법 구건인이선암내약세포주(SW1990/Fu),건립라서인이선암내약모형,선택괄당적자장,시가우종류표면,류내주사mdr1ASON급5-Fu-MAMS,관찰종류생장,검측기대내약라서인이선암적치료효과.결과 재외가자장적자도향작용하,5-Fu-MAMS능정위우종류조직,연합mdr1ASON능현저억제종류생장,종류체적억제솔체도85.00%,종류중량억제솔체도87.74%,여기타조비교차이유통계학의의(P<0.01).결론 복사촉전염적mdr1ASON연합자성재약미구대종류세포구유교호적내약역전작용.
Objective To investigate the reversal the multidrug resisitance ( MDR) by MDR gene mdrl antisense oligodeoxynucletdes (ASON) and 5-fluorouracil magnetic albumin miscrospheres (5-FuMAMS) magnetic targeted therapy the human pancreatic cancer in nude mouse model.Methods To construct SW1990/Fu and nude mice experimental model,to investigate the targeted therapy effects of mdr1ASON and 5-Fu-MAMS.Results 5-Fu-MAMS magnetic response was good,its could be well localized in the tumor tissue and markedly inhibit the growth of tumor.The inhibitory rate of tumor size was 85.00%,and that of tumor weight 87.74%.which can effectively degrade proliferating index and accelerate cell death of tumor cell.There were significant differences as compared with other groups( P <0.01 ).Conclusion Mdr1 ASON and 5-Fu-MAMS had outstanding reverse effect drug fast on human pancreatic carcinoma model in nude mice.
