目的 通过动物模型研究膝关节骨性关节炎软骨中基质金属蛋白酶(MMP)-3及金属蛋白酶组织抑制剂(TIMP)-1的表达及其与膝关节骨性关节炎之间关系.方法 雌性中国家兔制作软骨损伤模型,术后2周、4周、6周处死动物,分为实验组(E组)、实验对照组(C组)和正常对照组(N组).对膝关节软骨进行大体形态观察并行HE组织切片、免疫组化染色观察.结果 实验组膝关节呈进行性骨性关节炎改变,组织学观察显示透明软骨进行性退变,胶原破坏、蛋白聚糖降解、软骨细胞变性坏死.免疫组化染色显示透明软骨内MMP-3、TIMP-1表达均明显升高.结论 膝关节骨性关节炎造成软骨所处生物力学和生化环境紊乱,软骨组织结构退变、功能丧失而进一步加重骨性关节炎病变.

Objective To investigate the MMP-3 and TIMP-1 level in cartilage of rabbits osteoarthritis knee and their relationship with osteoarthritis. Methods Female rabbits were divided into Experiment group(E),Experimental contrast group(C) and Normal contrast group(N). Cartilage injury model was used in this study,animals were executed 2,4,6 weeks after surgery. Cartilage samples were observed,HE stain and immunohistochemistry were performed. Results Obvious osteoarthritis change was found in experiment group,include hyaline cartilage degeneration,collagen destroyed,proteoglycan degradation,cartilage cell necrosis. MMP-3 and TIMP-1 level increasing obviously in hyaline cartilage. Conclusions Biomechanics and biochemistry environment was compromised in knee osteoarthritis,also cartilage structure degenerated,malfunction of knee can conversely aggravate the osteoarthritis.
