目的探讨细胞因子白细胞介素-6(IL-6)和肿瘤坏死因子(TNF)与骨质疏松发生的关系。方法以去势后雌性SD大鼠为模型,分去势组、假手术组和去势加利维爱组,共三组,对照研究骨密度(BMD)和骨髓微环境中IL-6和TNF的含量变化。结果骨髓单核细胞具有分泌TNF和IL-6作用;卵巢分泌性激素水平下降可导致骨髓单核细胞TNF和IL-6基因表达调节失控;去势后大鼠体内TNF和IL-6水平与BMD呈负相关。结论提示TNF和IL-6的水平升高是引起骨丢失的机理之一,卵巢分泌性激素对TNF和IL-6具有下调作用。

Objective To study the relation between cytokines and osteoporosis.Methods In the present study the changes of bone mineral density (BMD)and interleukin-6(IL-6),tumor necrosis factor (TNF)concentration in bone marrow microenvironment were observed.We used ovariected female SD rats as osteoporosis model.The rats were randomly divided into three groups:ovariectomy (OVX),Sham-operation (Sham),OVX+Livial.Results 1.Bone marrow monocytes (BMM) secreted TNF,IL-6;2.The abnormal gene expression of TNF,IL-6 in BMM was resulted from the reduction in estrogen secreted by ovaries.Conclusion The increase of TNF,IL-6 concentration may be one of the pathogenesis of bone loss.Estrogen secreted by ovaries reduces TNF,IL-6 secretion in bone marrow microenvironment.
