目的:在离体鼠心Langendorff灌注模型上,检验并比较HTK液和UW液在低温(0~4℃)心肌保存中的心肌保护作用。方法:实验分为2组,HTK组和UW组,每组8只wistar大白鼠。快速取下鼠心并悬挂在Langendorff灌注模型上灌注和测定血流动力学基础值。灌注并低温保存离体鼠心4h。4h后重新复温复灌,再次测定血流动力学值,留取标本测定心肌水含量、灌脉流出液心肌酶漏出量、心肌中ATP和CP含量和观察心肌细胞超微结构变化。结果:心肌血流动力学恢复率HTK组优于UW组。心肌水含量两组间差异无显著意义。灌脉流出液心肌酶漏出量HTK组明显低于UW组。心肌中ATP和CP含量HTK组高于UW组。心肌细胞超微结构变化HTK组轻于UW组。结论:HTK液和UW液都是比较合适的心肌保存液,对心肌的保存效果HTK液优于UW液。

Objective: To compare the Histidine-Tryptophan-Ketogluarate(HTK) solution with that of University of Wisconsin (UW) solution in their heart preservation in an isolated rat heart preparation followig 4 hours hypothermia preservation. Method: The hearts isolated from male wistar rats were mounted on a langendorff apparatus to estimate baseline hemodynamic values. The hearts were divided into two groups (n=8 per group): HTK solution group and UW solution group. They were arrested and stored at 4℃ for 4 hours in each preservation solution. After that the hearts were reperfused and the recovery of myocardial function was evaluated again. Myocardial tissue water content, Myocardial tissue Adenosine Triphosphate (ATP) and Creatine Phosphate (CP) content in each group were measured. The leakage of myocardial enzymes was also finally, myocardial ultrastructure was observed. Result: The recovery rate of myocardial function: HTK group was significantly better than that in UE group (P<0.05). The levels of ATP and CP in myocardial tissue: HTK group was significantly better than that in UW group (P<0.01). The myocardial tissue water content: There was no statistical difference between HTK and UW groups. The leakage of myocardial enzymes: HTK group was significantly better than that in UW group (P<0.05); The myocardial ultrastructure: The histological changes of myocardial cell and the damage to the vascular endothelial cell were more significant in UW solution group compared with those in HTK solution group. Conclusion: These results suggest that both HTK solution and UW solution are fited for myocardial preservation with HTK solution more effective than UW solution.
