目的 筛选和鉴定Ⅰ期非小细胞肺癌(NSCLC)的血清差异表达蛋白.方法 分别收集经手术、病理证实的6例Ⅰ期NSCLC患者、7例肺部良性疾病患者和15例健康体检者的血清,利用双向凝胶电泳(2-DE)和基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)对Ⅰ期NSCLC的差异表达蛋白进行分离、筛选和鉴定.结果 每一标本的3张2-DE图谱半均蛋白质点数约(323±11)个.共挖取51个差异蛋白质点,经MALDI-TOF-MS分析,成功鉴定出5种蛋白质,均为上调蛋白,分别为α1-酸性糖蛋白、C1酯酶抑制物、血管紧张素原、转铁蛋白及转甲状腺素蛋白A链.结论 应用蛋白质组学技术在Ⅰ期NSCLC血清中鉴定出5种差异表达蛋白,为开发NSCLC早期诊断标志物提供了重要的候选蛋白.

Objective To identify differentially expressed proteins related to stage Ⅰ non-small cell lung cancer.Methods Sera were selected from 6 patients with stage Ⅰ non-small cell lung cancer,7 patients with benign lung disease confirmed surgically and pathologically,and 15 healthy volunteers.Two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) were employed to screen and identify differentially expressed serum proteins between lung cancer group and the other two groups.Results The average protein spots were (323 ±11 ). Ninety differentially expressed protein spots between lung cancer group and the other two groups were screened with PDQuest software,including 67 overexpressed protein spots and 25 underexpressed protein spots.Fifty significantly differentially expressed protein spots were excavated and 5 proteins were successfully identified via peptide mass fingerprinting using MALDI-TOF-MS,and all of them were overexpressed in the sera of lung cancer group,including Alpha-1-acid glycoprotein,C1 inbibitor,angiotensinogen,transferrin chain A and transthyretin.Conclusion Five overexpressed proteins can be identified in the sera of stage Ⅰ non-small cell lung cancer using 2-DE and MALDI-TOF-MS,which may serve as important candidate biomarkers for early detection of non-small cell lung cancer.
