目的:建立一种快速、高效的羊绒羊毛纺织品DNA提取的方法.方法:采用chelex-100法的3种处理、试剂盒法分别提取羊绒羊毛织品的DNA,用18S rDNA片段、山羊和绵羊源性成分PCR扩增结果来比较提取效果.结果:试剂盒法提取DNA的效果优于chelex-100法,整个提取过程约需2h.9种供试材料均提取到DNA,且含有山羊和,或绵羊源性成分,与显微镜观察结果的符合率为100%.结论:建立的试剂盒法是一种快速、高效的适用于羊绒羊毛织品DNA提取的方法,为应用分子生物学方法鉴别山羊绒和绵羊毛奠定了基础.

Objective:To develop a rapid and high effective DNA extraction method for cashmere and wool textiles.Method:With three treatments of chelex-100 method and kit method,the DNAs of cashmere and wool textiles were extracted.The DNA extraction effect of the two methods were compared by PCR expanding results of 18S rDNA segment,goat and sheep derived material.Result:The comparing results showed that the DNA extraction effect of kit method was better than that of chelex-100 method.It took about 2 hours to extract DNA by kit method.The DNAs of all nine experimental materials were extracted successfully,and all had goat and/or sheep derived material.The coincidence rate was 100% between the PCR detection results and the microscope results of these cashmere and wool textiles.Conclusion:The kit method developed was a rapid and high effective DNA extraction method for cashmere and wool textiles,which setting up a base for identification of cashmere and wool with molecule biology technique.
