目的 探讨臭氧氧化预处理通过诱导热休克蛋白70(HSP70)的合成,保护大鼠肾脏缺血再灌注损伤的作用与机制.方法 建立原位大鼠单侧肾缺血再灌注动物模型,I/R前15 d经直肠吹入氧气和臭氧的混合气体5.0~5.5 ml(臭氧浓度50 mg/L,1 mg/kg体蕈,每日 1次).全自动生化分析仪检测尿素氮(BUN)、肌酐(Cr),比色法测定血清的脂质过氧化产物丙二醛(MDA)、超氧化物歧化酶(SOD).Western blot检测HSP70蛋白的含量;逆转录聚合酶链反应(RT-PCR)方法检测HSP70的表达.结果 肾缺血再灌注24 h后,血清中BUN、Cr、MDA明显增高,肾组织内HSPT0表达明显增强(P<0.05),经臭氧氧化预处理后,血清中的BUN、Cr、MDA均降低,SOD升高;HSP70表达升高更加明显(P<0.05).结论 臭氧氧化预处理可以诱导大鼠肾缺血再灌注组织中HSP70表达,减轻大鼠肾脏缺血再灌注损伤.

목적 탐토취양양화예처리통과유도열휴극단백70(HSP70)적합성,보호대서신장결혈재관주손상적작용여궤제.방법 건립원위대서단측신결혈재관주동물모형,I/R전15 d경직장취입양기화취양적혼합기체5.0~5.5 ml(취양농도50 mg/L,1 mg/kg체심,매일 1차).전자동생화분석의검측뇨소담(BUN)、기항(Cr),비색법측정혈청적지질과양화산물병이철(MDA)、초양화물기화매(SOD).Western blot검측HSP70단백적함량;역전록취합매련반응(RT-PCR)방법검측HSP70적표체.결과 신결혈재관주24 h후,혈청중BUN、Cr、MDA명현증고,신조직내HSPT0표체명현증강(P<0.05),경취양양화예처리후,혈청중적BUN、Cr、MDA균강저,SOD승고;HSP70표체승고경가명현(P<0.05).결론 취양양화예처리가이유도대서신결혈재관주조직중HSP70표체,감경대서신장결혈재관주손상.
Objective To investigate the protective effect of ozone oxidative preconditioning (OzoneOP) on heat shock protein 70 (HSPT0) expression in renal ischemia-reperfusion (I/R) injury and the mechanisms. Methods A rat I/R model was established, and the renal OzoneOP was induced by rectal insufflation,1 mg/kg, 15 ozone treatments, once every day,5-5.5 ml at an ozone concentration of 50 mg/L. The parameters of renal function were determined by auto-biochemical analyzer. The contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured by using chromometry. HSP70 was examined by RT-PCR and Western blot. Results Compared with the sham group, the expression of HSP70 in renal tissues and the serum levels of BUN, CR and MDA were increased significantly in the rats undergoing I/R procedure. After OzoneOP, the levels of serum BUN, Cr, MDA were significandy decreased ,and SOD and HSP'/0 were significantly increased. Conclusion OzoneOP can increase the HSPTO expression and alleviate kidney I/R injury in rats.
