目的 观察胚胎干细胞和胰腺相关干细胞标志物在胰腺癌细胞系中基因和蛋白水平的表达,探讨胰腺癌起源.方法 应用逆转录聚合酶链反应(RT-PCR)检测胚胎干细胞标志物Oct4、abc2、c-kit以及c-kit配体干细胞因子(SCF)和胰腺相关干细胞标志物角蛋白19(ck19)、巢蛋白(nestin)、波形蛋白(vimentin)、胰十二指肠同源盒基因-1(PDX-1)等在胰腺痛细胞系sw1990、BxPC3、pc3和jt305中基因水平(mRNA)的表达;利用免疫细胞化学(ICC)方法检测Oct4、abcs2、c-kit和ck19、nestin在上述细胞系中蛋白水平的表达.同时结合异硫氰荧光素(FITC)标记抗体进行流式分选检测上述指标在胰腺癌细胞中的表达率.结果 除BxPC3、pc3和j1305不表达c-kit以外,上述4种胰腺癌细胞系在mRNA水平表达其余所有的标志物;在蛋白水平,上述4种细胞均表达Oct4、abcg2、ck19、nestin,而除sw1990以外的3株胰腺癌细胞株小表达c-kit.结论 人胰腺癌细胞系表达胚胎干细胞标志物Oct4、abcg2和胰腺干细胞标志物ck19、nestin、vimentin、PDX-1,部分表达c-kit.胰腺癌可能来源于成体胰腺干细胞或胰腺前体细胞.

목적 관찰배태간세포화이선상관간세포표지물재이선암세포계중기인화단백수평적표체,탐토이선암기원.방법 응용역전록취합매련반응(RT-PCR)검측배태간세포표지물Oct4、abc2、c-kit이급c-kit배체간세포인자(SCF)화이선상관간세포표지물각단백19(ck19)、소단백(nestin)、파형단백(vimentin)、이십이지장동원합기인-1(PDX-1)등재이선통세포계sw1990、BxPC3、pc3화jt305중기인수평(mRNA)적표체;이용면역세포화학(ICC)방법검측Oct4、abcs2、c-kit화ck19、nestin재상술세포계중단백수평적표체.동시결합이류청형광소(FITC)표기항체진행류식분선검측상술지표재이선암세포중적표체솔.결과 제BxPC3、pc3화j1305불표체c-kit이외,상술4충이선암세포계재mRNA수평표체기여소유적표지물;재단백수평,상술4충세포균표체Oct4、abcg2、ck19、nestin,이제sw1990이외적3주이선암세포주소표체c-kit.결론 인이선암세포계표체배태간세포표지물Oct4、abcg2화이선간세포표지물ck19、nestin、vimentin、PDX-1,부분표체c-kit.이선암가능래원우성체이선간세포혹이선전체세포.
Objective To investigate the expression of markers of human pancreas stem cells in four pancreatic cancer cell lines and study the origination of pancreatic cancer. Methods RT-PCR was used to detect mRNA expression of Oct4, c-kit, scf, ck 19, nestin, vimentin, abcg2, PDX-1, and immunocytochemistry was used to detect protein expression of Oct4,c-kit,ckl9, nestin,abcg2 in four pancreatic cancer cell lines including sw1990, BxPC3, pc3 and jf305. FITC-marked flow cytomytry was used to detect the expression rate of them in four pancreatic cancer lines. Results Four pancreatic cancer cell lines expressed mRNA of all Oct4,scf,ck19,nestin,vimentin,abcg2 and PDX-1. sw1990 expressed mRNA and protein of c-kit,but BxPC3,pc3 and jf305 did not express mRNA and protein of c-kit. Four pancreatic cancer cell lines expressed protein of Oct4,abcg2,ck19 and nestin. Conclusion Human pancreatic cancer cell lines express markers of human embryonic stem cells including Oct4, abcg2, scf and markers of pancreas stem cells including ck19, nestin, vimentin and PDX-1. Some pancreatic cancer cell lines do not expression ckit. Pancreatic cancer can originate from pancreas stem cells or precursor-cells of pancreas.
