筛选和鉴定与多囊卵巢综合征相关蛋白.方法:以固相pH梯度等电聚焦为第一向,垂直平板十二烷基磺酸钠聚丙烯酰胺凝胶电泳为第二向进行血清蛋白质分离;图像分析软件PDQuest 7.3分析电泳图谱寻找有意义的差异蛋白点,运用MALDI-TOF-MS质谱鉴定,分析其意义.结果:选取具有明显差异的3个蛋白点进行质谱鉴定,经数据库查询,2个蛋白点获得了有意义的结果分别为载脂蛋白A-I(Apolipoprotein A-I,ApoA-I),热休克蛋白70(Heat shock protein,HSP70).结论:多囊卵巢综合征患者与正常妇女的血清蛋白表达存在差异,这些差异蛋白有可能是多囊卵巢综合征特异相关性蛋白,并有可能成为多囊卵巢综合征诊断的分子标志物.

Objective: To find out disease specific-proteins (DSPs) which are closely collelated with Polycystic ovary syndrome. Methods: proteins ext racted from serum were run in immobilized pH gradient isoelectic focusing (IPG-IEF) at the first dimension.The proteins pots in gels were visuali zed by silver staining protocol followed by gel scanning and analysised by Pdquest 7.3 software. The differentially displayed protein spot s were searched and identifiyed by MALDI-TOF-MS. spots associated with PCOS. Rasults: Two DSPs were found and they were apolipoprotein A-I,Heat shock protein 70.Conclusion:There were different protein expressing in 2D-PAGE of Polycystic ovary syndrome and control group.These proteins may be DSPs of Polycystic ovary syndrome.
