目的探讨大鼠心肌缺血-再灌注时能量代谢及脂质过氧化变化及复方丹参滴丸的保护作用.方法采用Langendorff离体心脏灌流技术,制备心肌缺血再灌注损伤模型,分别在缺血前预灌时及缺血后再灌注时用复方丹参滴丸对心肌给予保护,观察心肌能量代谢及脂质过氧化(LPO)变化.本实验共分4组,正常对照组,单纯缺血再灌注组,复方丹参滴丸前保护组,复方丹参滴丸后保护组.结果①心肌缺血40min再灌注20min,心肌组织内高能磷酸化合物明显减少,脂质过氧化物含量明显增多,与正常对照组比P<0.01.②在缺血前预灌注时及缺血后再灌注时加入复方丹参滴丸,心肌组织中的高能磷酸化合物均高于单纯缺血再灌注组(P<0.01),两组ATP、TAN均接近正常水平(P>0.05).两组的LPO低于单纯缺血再灌注组,复方丹参滴丸前保护组的LPO与对照组比无明显差别(P>0.05).结论心肌缺血40min再灌注20min,心肌能量代谢障碍,脂质过氧化反应增强,大鼠心肌发生缺血再灌注损伤.复方丹参滴丸在缺血前预灌注时与缺血后再灌注时给予均可通过增加心肌能量储备,抑制脂质过氧化物的生成而保护心肌.

목적탐토대서심기결혈-재관주시능량대사급지질과양화변화급복방단삼적환적보호작용.방법채용Langendorff리체심장관류기술,제비심기결혈재관주손상모형,분별재결혈전예관시급결혈후재관주시용복방단삼적환대심기급여보호,관찰심기능량대사급지질과양화(LPO)변화.본실험공분4조,정상대조조,단순결혈재관주조,복방단삼적환전보호조,복방단삼적환후보호조.결과①심기결혈40min재관주20min,심기조직내고능린산화합물명현감소,지질과양화물함량명현증다,여정상대조조비P<0.01.②재결혈전예관주시급결혈후재관주시가입복방단삼적환,심기조직중적고능린산화합물균고우단순결혈재관주조(P<0.01),량조ATP、TAN균접근정상수평(P>0.05).량조적LPO저우단순결혈재관주조,복방단삼적환전보호조적LPO여대조조비무명현차별(P>0.05).결론심기결혈40min재관주20min,심기능량대사장애,지질과양화반응증강,대서심기발생결혈재관주손상.복방단삼적환재결혈전예관주시여결혈후재관주시급여균가통과증가심기능량저비,억제지질과양화물적생성이보호심기.
Objective To study the energy metabolism and lipid peroxidation in rat myocardium with ischemia/reperfusion and the protective effect of compound Danshen dripping pill(DSDP).Methods The model of myocardium with ischemia/reperfusion was made in isolated rat heart perfused with Langendorff apparatus.Compound DSDP was given to myocardium in preperfusion before ischemia and in reperfusion after ischemia,to observe the changes of energy-metabolism and lipid peroxide(LPO) in myocardium.There were four groups in the test,that is,control group,IR group,pre-DS group and post-DS group.Results ①In the time of 40 minute myocardial ischemia and 20 minute re-perfusion,there was a remarkable decrease in high-energy phoshphate and a distinct increase in lipid peroxide in myocardium,compared with those in control group(P<0.01).②When DSDP was given in preperfusion before and after ischemia,high-energy phosphate was higher than that in only ischemia/reperfusion group(P<0.01).In both groups,ATP and total adenosine pool (TAN) were close to normal(P>0.05),and LPO was lower than that in IR group(P<0.01).LPO in pre-DS group was not significantly different from that in control group(P>0.05).Conclusion When myocardium was ischemined for 40min then reperfused for 20min,high energy phosphate synthesis decreased and lipid peroxidation increased,and myocardium suffered from ischemia/reperfusion injury.Either given in pre-perfusion before ischemia and post-perfusion after ischemia,DSDP could increase the energy storage of myocardium and protect it by inhibiting the generation of lipid peroxide.So DSDP could be used in prevention and treatment of ischemia reperfusion injury.
