目的用糖耐量曲线下面积(SBG)、胰岛素释放曲线下面积(SIns)、胰岛素敏感指数,这些临床糖尿病常用的指标对非胰岛素依赖型糖尿病(non insulin-dependent diabetes mellitus,NIDDM)模型胰岛素抵抗评估。方法大鼠尾静脉注射小剂量链尿佐菌素,使大鼠糖耐量异常,然后加喂高热量食物,引起动物肥胖,饲养8wk,可形成类似NIDDM的肥胖大鼠模型,并设置对照组,同时测定了大鼠糖耐量试验过程中血清葡萄糖(BG)、胰岛素(Ins),并计算了SBG、SIns、胰岛素敏感指数。结果①模型组糖耐量试验,血清BG糖耐量下降,Ins在糖负荷后2h明显升高(P<0.01);②Ins敏感指数较对照组明显降低(P<0.01);③SBG明显上升(P<0.01);SIns无明显变化,SIns/SBG下降(P<0.01)。结论 SBG、SIns、胰岛素敏感指数可较好反映Ins作用,B细胞储备功能及Ins敏感性,可广泛用于实验性糖尿病基础研究和作为临床正确评价IR的指标。
目的用糖耐量麯線下麵積(SBG)、胰島素釋放麯線下麵積(SIns)、胰島素敏感指數,這些臨床糖尿病常用的指標對非胰島素依賴型糖尿病(non insulin-dependent diabetes mellitus,NIDDM)模型胰島素牴抗評估。方法大鼠尾靜脈註射小劑量鏈尿佐菌素,使大鼠糖耐量異常,然後加餵高熱量食物,引起動物肥胖,飼養8wk,可形成類似NIDDM的肥胖大鼠模型,併設置對照組,同時測定瞭大鼠糖耐量試驗過程中血清葡萄糖(BG)、胰島素(Ins),併計算瞭SBG、SIns、胰島素敏感指數。結果①模型組糖耐量試驗,血清BG糖耐量下降,Ins在糖負荷後2h明顯升高(P<0.01);②Ins敏感指數較對照組明顯降低(P<0.01);③SBG明顯上升(P<0.01);SIns無明顯變化,SIns/SBG下降(P<0.01)。結論 SBG、SIns、胰島素敏感指數可較好反映Ins作用,B細胞儲備功能及Ins敏感性,可廣汎用于實驗性糖尿病基礎研究和作為臨床正確評價IR的指標。
목적용당내량곡선하면적(SBG)、이도소석방곡선하면적(SIns)、이도소민감지수,저사림상당뇨병상용적지표대비이도소의뢰형당뇨병(non insulin-dependent diabetes mellitus,NIDDM)모형이도소저항평고。방법대서미정맥주사소제량련뇨좌균소,사대서당내량이상,연후가위고열량식물,인기동물비반,사양8wk,가형성유사NIDDM적비반대서모형,병설치대조조,동시측정료대서당내량시험과정중혈청포도당(BG)、이도소(Ins),병계산료SBG、SIns、이도소민감지수。결과①모형조당내량시험,혈청BG당내량하강,Ins재당부하후2h명현승고(P<0.01);②Ins민감지수교대조조명현강저(P<0.01);③SBG명현상승(P<0.01);SIns무명현변화,SIns/SBG하강(P<0.01)。결론 SBG、SIns、이도소민감지수가교호반영Ins작용,B세포저비공능급Ins민감성,가엄범용우실험성당뇨병기출연구화작위림상정학평개IR적지표。
Objective To analyse insulin resist (IR) of non insulin-dependent diabetic rat model with insulin sensitive index,the area of and glucose under curve (SBG),the area of insulin under curve (SIns),SIns/SBG which were used in diabetic patient.Methods Rats were injected with small dose streptozocoi (iv).Two weeks after the injection,the rats developed impaired glucose tolerance.Then,they were fed with high energy diet for eight weeks to form non-insulin dependent diabetes mellitus (NIDDM).Concentrations of blood glucose and serum insulin in oral glucose tolerance were detected.SBG,Sins which is used describing IR and function of pancreas β cell are also calculated.Results ①The glucose tolerance was impaired,hyper insulin was found in the oral glucose tolerance test(P<0.01).②Insulin sensitive index decreased compared with controls (P<0.01).③SBG was significantly higher than that of control (P<0.01),but no significant difference was found with SIns between two groups.SIns/SBG decreased compared with controls(P<0.01).Conclusions Insulin sensitive index、SBG、SIns/SBG may reflect affect of insulin、function of pancreas β cell and insulin sensityvity,which were widely applied to basic research of experimental diabetes,or as evaluated index of IR.
