研究环孢菌素A(CsA)对肺纤维化的治疗作用。方法:应用博莱霉素(BLM)复制大鼠
肺纤维化模型,观察CsA对支气管肺泡灌洗液(BALF)中细胞成分、肺组织学变化以及应用SABC法检测肺组织中纤维连接蛋白(FN)和血小板衍化生长因子(PDGF)表达的作用。结果:应用博莱霉素后第7天即有严重肺泡炎性改变,晚期有严重的肺纤维化,而CsA组,早期只有轻度炎性改变,BALF中细胞计数较BLM组低(P<0.001),FN及PDGF表达显著弱于BLM组(P<0.01)。结论:CsA通过抑制FN及PDGF的表达治疗肺纤维化。

폐섬유화모형,관찰CsA대지기관폐포관세액(BALF)중세포성분、폐조직학변화이급응용SABC법검측폐조직중섬유련접단백(FN)화혈소판연화생장인자(PDGF)표체적작용。결과:응용박래매소후제7천즉유엄중폐포염성개변,만기유엄중적폐섬유화,이CsA조,조기지유경도염성개변,BALF중세포계수교BLM조저(P<0.001),FN급PDGF표체현저약우BLM조(P<0.01)。결론:CsA통과억제FN급PDGF적표체치료폐섬유화。
To study the effects of cyclosporin A on pulmonary fibrosis induced by bleomycin in rats. Methods: Forty five wistar rats were divided into three groups in random: Saline, untreated model and cy
closporin A-treated group. Bronchoalveolar lavage were performed in these rats for the enumeration of cells. Lung tissues of tested animals were pathologically observed and immunohistochemically stained. Results: Seven days later after using bleomycin instillation,Serious alveolitis presented,and the serious pulmonary fibrosis formed at 28th day. In contrast to the bleomycin group, in the early course of the cyclosporin A-treated group, there was
mild inflammatory changes observed under microscope. The expressions of FN and PDGF decreased more in the
cyclosporin A-treated group compared with those of the same stage in the bleomycin group, There was almost disappearance of pulmonary fibrosis in the cyclosporin A-treated group at 28th day. Conclusion: Cyclosporin A may have an inhibitory effect on the process of the pulmonary fibrosis.
