目的:观察8-羟基脱氧鸟苷(8-hydroxy-deoxyguanosine,8-OHdG)在2型糖尿病(Diabetes mellitus,DM)模型大鼠肾脏的表达及普罗布考(Probucol)对它的影响.方法:将大鼠分为正常对照组(Normol rats,C)、DM组(Diabetic rats,D)、普罗布考治疗组(Diabetic rats treated with probucol,N),比较各组大鼠肾组织中丙二醛(Malondialdehyde,MDA)含量、超氧化物岐化酶(Superoxide dismutase,SOD)活性,和8羟基脱氧鸟苷(8-OHdG)含量,同时观察血糖、血胰岛素、肾功能和肾脏形态变化.结果:与正常组比较,DM组血液中MDA含量明显上升(P<0.05),SOD活性显著下降(P<0.05),8-OHdO增多(P<0.05),肾功能和肾脏形态受损,而普罗布考治疗组的上述表达较DM组明显改善.结论:8-OHdG在DM肾病(Diabeticnephropathy,DN)中的表达明显增加,而普罗布考可以通过抑制氧化应激反应对2型DM模型大鼠肾脏产生保护作用.

목적:관찰8-간기탈양조감(8-hydroxy-deoxyguanosine,8-OHdG)재2형당뇨병(Diabetes mellitus,DM)모형대서신장적표체급보라포고(Probucol)대타적영향.방법:장대서분위정상대조조(Normol rats,C)、DM조(Diabetic rats,D)、보라포고치료조(Diabetic rats treated with probucol,N),비교각조대서신조직중병이철(Malondialdehyde,MDA)함량、초양화물기화매(Superoxide dismutase,SOD)활성,화8간기탈양조감(8-OHdG)함량,동시관찰혈당、혈이도소、신공능화신장형태변화.결과:여정상조비교,DM조혈액중MDA함량명현상승(P<0.05),SOD활성현저하강(P<0.05),8-OHdO증다(P<0.05),신공능화신장형태수손,이보라포고치료조적상술표체교DM조명현개선.결론:8-OHdG재DM신병(Diabeticnephropathy,DN)중적표체명현증가,이보라포고가이통과억제양화응격반응대2형DM모형대서신장산생보호작용.
Objective:To evaluate the expression of 8-hydroxyguanine(8-OHdG ) in type 2 diabetic rats and the effect of Probucol on it. Methods:25 rats were divided into 3 groups of normal rats(C), diabetic rats (D), and diabetic rats treated with Probucol(N). Content of malonaldehyde (MDA) and activity of antioxidant enzymes including superoxid edismutase (SOD) and 8-OHdG in the renal tissue were compared among the groups. In addition,blood glucose,serum creatinine (Cr) and blood ureanitrogen (BUN)were mesured;the renal tissue were observed by light micros copy and electron microscopy. Results:Compared with normal group,the level of BUN,Cr, MDA and 8-OHdG in Probucol untreated group were significantly increased,but the activity of SOD were decreased notably. In addition,the renal pathologic changes in Probucol untreated group was also decreased, but improved in the treared group obviously.Conclusion: Oxidative stress takes key point in the development of diabetic nephropathy. The expression of 8-OHdG in type2 diabetic rats were increased. Andantioxidation may be the possible mechanism of the nephroprotective action of Probucol.
