目的 为获取中华按蚊感染约氏疟原虫后24 h的高表达基因热激蛋白40(HSP40)cDNA的全长序列. 方法 运用抑制性消减杂交技术(suppression substractive hybridization,SSH)构建中华按蚊感染约氏疟原虫后24 h的差异表达基因文库,从高表达文库中获得长度为363 bp的HSP40基因的cDNA片段,运用RACE技术对其cDNA序列进行全长扩增. 结果 获得中华按蚊HSP40 cDNA的全长序列,总长度为1 159 bp,开放阅读框为1 014 bp,编码337个氨基酸. 结论 获得了中华按蚊HSP40 cDNA的全长序列.

Objective To isolate differentially expressed gene HSP40 from Anopheles sinensis induced by infection with Plasmodium yoelii yoelii after 24 h, and to amplify the full-length cDNA of HSP40. Methods A 363 bp of HSP40 cDNA was isolated from the suppression substractive hybridization (SSH) library. The full length of cDNA was amplified using the RACE (rapid-amplification of cDNA ends) technology. Results The full-length cDNA of HSP40 was completely amplified. The whole length was 1 159 bp, with an open reading frame of 1 014 bp, encoding 337 amino acids. Conclusions The full-length cDNA of HSP40 has been completely amplified.
