目的评估HCCR1和HCRR2克隆、细菌表达及可溶性蛋白的纯化及应用于早期肝癌诊断的可行性.方法cDNA克隆、表达、蛋白质的纯化、Western blot分析用于评估HCCR1和HCCR2蛋白的生产,Elisa方法用检测血清抗原.结果HCCR1和HCCR2能够高水平地在细菌中表达,纯化的可溶性蛋白能用于生产抗体并应用于肝癌的早期诊断.结论HCCR1和HCCR2蛋白质可以在细菌中生产,并能成功地用于肝癌的早期诊断.
目的評估HCCR1和HCRR2剋隆、細菌錶達及可溶性蛋白的純化及應用于早期肝癌診斷的可行性.方法cDNA剋隆、錶達、蛋白質的純化、Western blot分析用于評估HCCR1和HCCR2蛋白的生產,Elisa方法用檢測血清抗原.結果HCCR1和HCCR2能夠高水平地在細菌中錶達,純化的可溶性蛋白能用于生產抗體併應用于肝癌的早期診斷.結論HCCR1和HCCR2蛋白質可以在細菌中生產,併能成功地用于肝癌的早期診斷.

[Objective] HCCR1 and HCCR2 are the oncogenes overexpressed in hepatocellular carcinomas(HCC). In this experiment, it was evaluated that the cloning, bacterial expression, purification and characterization of soluble recombinant proteins of HCCR1 and HCCR2 for novel early diagnosis of HCC. Protein solublity was key to producing recombinant proteins for founction determination. [Method] Five methods, cDNA clone and expression,protein purification, solubility assay and western blod assay were used to evaluate production of HCCR1 and HCCR2 proteins, enzyme-linked immunosorbent assay (ELISA) was used to detect HCCR1 and HCCR2 antigens in serum.[Result] The results show that the proteins of HCCR1 and HCCR2 were expressed in Escherichia coli at high levels. [Conclusion] The recombinant proteins are soluble and may used to generate the antibody for early diagnosis of HCC. Finally, HCCR1 and HCCR2 may be useful in monitoring HCC, and as potential disease specific therapeutic targets.
