目的探讨大肠组织中基质金属蛋白酶-2(MMP-2)和T淋巴瘤侵袭转移诱导因子1(Tiam1)的基因表达与大肠癌生物学特征的相关性.方法应用S-P免疫组化法对46例大肠癌手术切除标本进行抗人MMP-2单克隆抗体和Tiaml1多克隆抗体免疫组化染色,检测癌组织、癌旁组织、手术切缘区正常组织各区域MMP-2和Tiam1 蛋白表达,分析二者与大肠癌临床病理特征之间的关系.结果 MMP-2和Tiam1蛋白表达在癌组织与手术切缘区正常组织、癌组织与癌旁组织、癌旁组织与手术切缘区正常组织之间差异均具有统计学意义.癌组织中MMP-2和Tiam1蛋白表达在患者性别、年龄之间差异无统计学意义,而与肿瘤分化程度、浸润深度、淋巴结及远处转移、临床TNM分期均呈正相关；且两种蛋白表达同时阳性的大肠癌病例具有更高恶性的生物学特征.结论 MMP-2和Tiam1与大肠癌的发生、侵袭和转移具有潜在影响作用,检测MMP-2和Tiam1的表达可作为判断大肠癌病变发展和转移潜能的指标.

Objective To study the relationship between the matrix metalloproteinase-2(MMP-2),T-lymphoma invasion and metastasis inducing factor 1 (Tiam1)and the biological characteristics of colorectal carcinoma.Methods A total of 46 resected specimens were investigated by S-P immunohistochemical staining with anti-MMP-2 monoclonal antibody and anti-Tiam1 polyclonal antibody.The expression of MMP-2 and Timal in various regions of the colorectal carcinomas tissues,paracarcinoma tissues and incisal margin normal tissues were detected and the relationship with clinicopathologic factors were analyzed.Results The difference of protein expresssion of MMP-2 and Tiaml in colorectal carcinomas tissues versus incisal margin normal tissues,colorectal carcinomas tissues versus paracarcinoma tissues,paracarcinoma tissues versus incisal margin normal tissues were statistically significant.The protein expressions of MMP-2 and Tiam1 in colorectal carcinomas tissues were not related with sex and age,but were positively related with the differention,depth of invasion,lymph node metastasis,distant metastasis and TNM stage,and there was a positive correlation between the MMP-2 expression and Tiam1 expression.The case of colorectal carcinoma was more malignant while both of the MMP-2 expression and Tiam1 expression were positive.Conclusions MMP-2 and Tima1 are closely related to carcinogenesis,invision and metastasis of colorectal carcinoma.The protein expressions of MMP-2 and Tiam1 could be served as the determinant factors for estimating tumor progress and predicting occurrence of carcinoma cell metastasis.
