应用特殊设计的寡核苷酸链纠正点突变技术是20世纪90年代发展起来的一项新技术,在酵母菌、植物细胞、哺乳动物细胞等不同的生物材料,甚至动物模型中都有越来越多成功的报道.该技术还处于探索阶段,其基因修复机制还不是很清楚,目前认为突变纠正率与寡核苷酸链的结构、转运体系、靶基因的转录、DNA的复制、错配修复、损伤修复等生物学过程有关.

The technology that correcting the point mutation by specially designed oligonucleotides is a laest molecular biological technique developed from late 1990s'. Successful repair has been achieved in yeast,plant, mammalian cells and other biological material, even in more and more animal models. But the technology is still in its developing and growing stage, and the gene repair mechanisms need to be elucidated more exactly.At present, the following factors are considered to be related; the structures and transport systems of oligonucleotides, transcription of the target gene, DNA replication, mismatch repair, damage repair and other relate biological processes.
