为测试复方黄连乳腺炎注射液以其主要有效成分小檗碱体内和体外抑菌效果.采用琼脂平板扩散法(打孔法)和试管二倍稀释法,用复方黄连乳腺炎注射液对引起奶牛乳房炎的3种主要致病菌(金黄色葡萄球菌、大肠杆菌、无乳链球菌)进行体外抑菌试验,结果显示对金黄色葡萄球菌抑菌圈直径为42 .3 mm,对大肠杆菌抑菌圈直径为35.2 mm,对无乳链球菌抑菌圈直径为41.5 mm.采用小鼠乳腺炎模型对小檗碱体内抑菌效果和对小鼠乳腺组织中细胞因子的影响进行试验,结果显示:与模型组相比,低剂量组、高剂量组在接菌12~48 h后小鼠乳腺组织中的细菌数均极显著低于模型组(P<0 01);低剂量组、高剂量组小鼠乳腺组织TNF-α在接菌24 h后极显著低于模型组(P<0 01);接菌24~36 h后,低剂量组和高剂量组乳腺组织IFN-γ含量极显著低于模型组(P<0 01).结果表明:复方黄连乳腺炎注射液对金黄色葡萄球菌、大肠杆菌、无乳链球菌均有较好抑菌效果,其主要有效成分小檗碱能明显抑制小鼠乳腺内金黄色葡萄球菌的增殖,降低乳腺组织TNF-α、IFN-γ水平,从而减轻炎症介质对乳腺的病理损伤.

위측시복방황련유선염주사액이기주요유효성분소벽감체내화체외억균효과.채용경지평판확산법(타공법)화시관이배희석법,용복방황련유선염주사액대인기내우유방염적3충주요치병균(금황색포도구균、대장간균、무유련구균)진행체외억균시험,결과현시대금황색포도구균억균권직경위42 .3 mm,대대장간균억균권직경위35.2 mm,대무유련구균억균권직경위41.5 mm.채용소서유선염모형대소벽감체내억균효과화대소서유선조직중세포인자적영향진행시험,결과현시:여모형조상비,저제량조、고제량조재접균12~48 h후소서유선조직중적세균수균겁현저저우모형조(P<0 01);저제량조、고제량조소서유선조직TNF-α재접균24 h후겁현저저우모형조(P<0 01);접균24~36 h후,저제량조화고제량조유선조직IFN-γ함량겁현저저우모형조(P<0 01).결과표명:복방황련유선염주사액대금황색포도구균、대장간균、무유련구균균유교호억균효과,기주요유효성분소벽감능명현억제소서유선내금황색포도구균적증식,강저유선조직TNF-α、IFN-γ수평,종이감경염증개질대유선적병리손상.
This study aims to test the bacteriostasis effect of compound Rhizoma coptidis injection on mastitis and Berberine of main active ingredient. By agar diffusion method (punch method) and two-fold dilution method in test tube,in vitro bacteriostasis test about Staphylococcus aureus,Escherichia coli,Streptococcus agalactiae of compound Rhizoma coptidis injection on mastitis were investigated. Results showed that the bacteriostasis diameters of compound Rhizoma coptidis injection on Staphylococcus aureus,E.coli,S. agalactiae were 42 3,35 2 and 41 5 mm respectively. With the mouse mastitis model induced by S. aureus,the bacteriostasis effect of berberine chloride in mammary gland and its inflammation-associated factors were investigated. Bacterial counts of low and high dose groups were lower significantly than that of model group in mammary gland after 12-48 h of inoculation(P<0 01),TNF-α level in mammary gland of low and high dose groups were lower significantly than that of model group after 24 h of inoculation(P<0 01),IFN-γ level in mammary gland of low and high dose groups were lower significantly than that of model group after 24-36 h of inoculation (P<0 01). Furthermore,bacteriostasis effect of compound R. coptidis injection on these three pathogens were very sensitive,berberine could inhibit S. aureus growth,and decrease TNF-α、IFN-γ level in mammary gland,inhibit excessive inflammatory reaction,alleviate damages of mammary gland from proinflammatory factors.
