目的 探讨骨组织经不同浓度的甲酸脱钙液24小时脱钙后切片染色效果,选择最佳甲酸脱钙工作液.方法 350例骨组织标本每例取材3份,分别用40%、30%、20%的甲酸脱钙液常温下脱钙24小时,观察三种不同浓度的甲酸脱钙液脱钙后骨组织的切片和HE染色情况.结果 骨组织经40%甲酸脱钙液脱钙后,切片完整,但HE染色核质偏浅,胞浆偏红;30%甲酸脱钙液脱钙后,切片完整,HE染色核质清晰,胞浆鲜艳;20%甲酸脱钙液脱钙后,切片不够完整,HE染色核质偏浅,胞浆偏浅.结论 脱钙液脱钙后应保持组织切片完整,染色效果良好,且30%的甲酸脱钙液是较理想的工作液.

목적 탐토골조직경불동농도적갑산탈개액24소시탈개후절편염색효과,선택최가갑산탈개공작액.방법 350례골조직표본매례취재3빈,분별용40%、30%、20%적갑산탈개액상온하탈개24소시,관찰삼충불동농도적갑산탈개액탈개후골조직적절편화HE염색정황.결과 골조직경40%갑산탈개액탈개후,절편완정,단HE염색핵질편천,포장편홍;30%갑산탈개액탈개후,절편완정,HE염색핵질청석,포장선염;20%갑산탈개액탈개후,절편불구완정,HE염색핵질편천,포장편천.결론 탈개액탈개후응보지조직절편완정,염색효과량호,차30%적갑산탈개액시교이상적공작액.
Objective This study is to investigate the decalcification effect of different concentrations of formic acid solution and to determine the best one.Methods Bone tissues from 330 cases were dissected and equally and randomly divided into 3 shares.Each was immersed in the formic acid of 40%,30% and 20% concentration for 24 h separately.Then the efficiency of cutting section and the HE stain were evaluated.Result With all three concentrations of formic acid solution,the sections were cut smoothly and completely in all the groups except the 20% concentration group.The HE stain of some cases from 20% concentration groups was pale in the stain of the nuclei and the cytoplasm.The HE stain of some cases from 40% concentration groups was pale in the stain of the nuclei and reddish in the cytoplasm.Both the nuclei and cytoplasm were stained well in the groups of 30% concentration.Conclusion The slides of bone tissue should be kept intact and well stained.So,in our study,the best choice is the formic acid of 30% concentration.
