目的研究清络通痹方(Qingluo - Tongbi Grannual,QLT)对类风湿关节炎(rheumatoid arthritis,RA)患者外周血T淋巴细胞表达RANKL( receptor activator of nuclear factor - KB ligand,破骨细胞分化因子)的影响.方法临床纳入15例RA患者,肘静脉抽血10ml,每个血样分为5组,包括正常对照组,阳性对照组,和QLT小、中、大三个剂量组.血样加药培养72小时后,加入FITC标记的CD3抗体和PE标记的RANKL抗体,采用流式细胞术检测RANKL的表达率.结果采用中剂量和大剂量QLT动物含药血清培养的T淋巴细胞RANKL表达率受到了显著的抑制,与正常对照组比较具有统计学差异(P＜0.01).结论 QLT可以有效下调RA患者外周血T淋巴细胞RANKL的表达.
목적 연구청락통비방(Qingluo - Tongbi Grannual,QLT)대류풍습관절염(rheumatoid arthritis,RA)환자외주혈T림파세포표체RANKL( receptor activator of nuclear factor - KB ligand,파골세포분화인자)적영향.방법 림상납입15례RA환자,주정맥추혈10ml,매개혈양분위5조,포괄정상대조조,양성대조조,화QLT소、중、대삼개제량조.혈양가약배양72소시후,가입FITC표기적CD3항체화PE표기적RANKL항체,채용류식세포술검측RANKL적표체솔.결과 채용중제량화대제량QLT동물함약혈청배양적T림파세포RANKL표체솔수도료현저적억제,여정상대조조비교구유통계학차이(P＜0.01).결론 QLT가이유효하조RA환자외주혈T림파세포RANKL적표체.
Objective To study the effect of medicated serum containing Qingluo - tongbi Granule (QLT) on RANKL ( receptor activator of nuclear factor - κB ligand) expression of T lymphocytes extracted from peripheral blood of rheumatoid arthritis (RA) patients.Methods Blood samples extracted from veins of 15 RA cases were divided into 5 groups,including normal control,positive control,small - dose QLT,medium - dose as well as large -dose.Samples were cultured,processed under general program and tested with flow cytometer after antibodies of FITC labeled anti -human CD3 and PE labeled anti -human RANKL were added in.Results Compared with normal control group,RANKL expression of T lymphocytes treated with serum containing medium or large dose QLT was inhibited obviously in vitro ( P ＜ 0.01 ),and QLT demonstrate effect to suppress RANKL expression.Conclusion QLT is able to suppress RANKL over - expression of T lymphocytes of RA patients.
