Rhizoma Anemarrhenae%Yin deficiency%Anxiety
目的 观察知母对阴虚小鼠焦虑行为的影响,筛选活性物质并探讨其机制.方法 采用320 mg/kg甲状腺片混悬液对小鼠连续灌胃9d造阴虚模型,以高架十字迷宫和明暗箱装置观察阴虚模型小鼠焦虑行为状态.行为学测试结束后,立即断头处死,取脑,左右脑分别测定GABA和Glu的含量以及GABAAR1和NMDAR1表达.结果 ①小鼠高架十字迷宫实验进入开臂的时间比和次数比,阴虚模型组[(28.83±7.5)%,(24.1±4.0)%,n=10]较地西泮组[(40.6±4.9)%,(33.8±4.2)%,n=10]和醇提物中剂量组[(40.4±0.4)%,(35.0±3.3)%,n=10]显著减少(P<0.05);在明暗箱实验中,阴虚模型组穿箱次数[(17.5±1.6)次,n=10]较地西泮组[(18.7±0.8)次,n=10]和醇提物中剂量组[(18.6±1.4)次,n=10]显著减少(P<0.05).②小鼠脑内GABA含量,阴虚模型组[(24.45±4.96)μg/g,n=10]较地西泮组[(46.59±11.85)μg/g,n=10]和醇提物中剂量组[(48.97±9.14)μg/g,n=10]显著降低(P<0.05);Glu含量,阴虚模型组[(440.6±108.8)μg/g,n=10]较地西泮组[(208.5±77.88)μg/g,n=10]和醇提物中剂量组[(255.6±77.01)μg/g,n=10]显著升高(P<0.05).③与阴虚模型组比较,地西泮组和醇提物中剂量组小鼠脑内GABAAR1平均积分光密度值显著升高(P<0.05)、NMDAR1平均积分光密度值显著降低(P<0.05).结论 知母醇提物具有明显的抗焦虑作用,其机制可能与提高小鼠脑内GABA含量和GABAAR1表达,降低Glu含量和NMDAR1表达有关.

Objective To observe the effects of Rhizoma Anemarrhenae on the anxiety behavior of the yin deficiency mice, to filter the active material of Spine date seed for yin deficiency model small mice anxiety behavior and to discuss the mechanism. Methods Yin deficiency model with intragastrical 320 mg/kg thyroid tablet suspention for nine days exception control group. Anxiety action was observed yin deficiency model small mice by the means of the elevated plus-maze and light-dark transitions of mice. Then executed it immediately, and peeled off cerebra at once, the left saved in liquid nitrogen for assay of GABA and Glu, and right saved in formalin for deterclose-arm ( OE% ) of the elevated plus-maze test : compared with the diszepam group [ (40.6±4.9 )% , ( 33.8±4.2 ) %, n = 10 ] and the middle alcohol extracts group [ (40.4±0.4 )%, ( 35.0±3.3 )%, n = 10 ] , yin deficiency model group[ ( 28.83±7.5 ) %, ( 24.1±4.0 )%, n = 10 ] obviously deficiency ( P < 0.05 ). The passing times in light-dark transitions test: compared with the diszepam group( 18.70±0.8, n = 10) and the middle alcohol extracts group( 18.6±1.4, n = 10 ), yin deficiency model group( 17.5±1.6, n = 10 ) obviously decrease ( P10] and the middle alcohol extracts group[ (48.97±9.14) μg/g, n= 10], yin deficiency model group[ (24.45±4.96)μg/g, n = 10 ] obviously decrease ( P < 0.05 ) ; and the level of Glu, compared with the diszepam group [ ( 208.5±77.88 ) μg/g, n = 10 ] and the middle alcohol extracts group[ (255.6±77.01 )μg/g, n = 10 ], yin deftciency model group, the expression of GABAAR1 increased evidently( P< 0.05 ) and NMDAR1 reduced evidently ( P < 0.05 ) of the diszepam group and the middle alcohol extracts group. Condusion The alcohol extracts of Rhizoma Anemarrhenae have the anti-anxiety effects instinctively. Its mechanism may be of relating to increasing the GABA and GABAAR1 and reducing the Glu and NMDAR1.
