目的:观察中药复方别敏胶囊对呼吸道变应性模型小鼠内分泌免疫功能的影响.方法:利用卵清蛋白建立呼吸道变应性小鼠模型,采用苏木精和伊红染色观察肺组织病理变化,运用流式细胞仪检测脾淋巴细胞中Th1和Th2细胞变化,利用酶联免疫吸附测定方法检测脾淋巴细胞培养上清白细胞介素4(interleukin-4, IL-4)、白细胞介素5(interleukin-5, IL-5)、干扰素γ(interferon-gamma, INF-γ)水平及血清皮质酮浓度.结果:别敏能够明显减轻模型小鼠肺组织的炎症反应,降低模型小鼠Th2细胞及其细胞因子IL-4、IL-5水平,升高Th1细胞及其细胞因子INF-γ水平,显著抑制模型小鼠内源性皮质酮水平的下降. 结论:别敏对机体内分泌免疫系统功能紊乱的调节作用可能是其治疗呼吸道变应性疾病的重要机制之一.
目的:觀察中藥複方彆敏膠囊對呼吸道變應性模型小鼠內分泌免疫功能的影響.方法:利用卵清蛋白建立呼吸道變應性小鼠模型,採用囌木精和伊紅染色觀察肺組織病理變化,運用流式細胞儀檢測脾淋巴細胞中Th1和Th2細胞變化,利用酶聯免疫吸附測定方法檢測脾淋巴細胞培養上清白細胞介素4(interleukin-4, IL-4)、白細胞介素5(interleukin-5, IL-5)、榦擾素γ(interferon-gamma, INF-γ)水平及血清皮質酮濃度.結果:彆敏能夠明顯減輕模型小鼠肺組織的炎癥反應,降低模型小鼠Th2細胞及其細胞因子IL-4、IL-5水平,升高Th1細胞及其細胞因子INF-γ水平,顯著抑製模型小鼠內源性皮質酮水平的下降. 結論:彆敏對機體內分泌免疫繫統功能紊亂的調節作用可能是其治療呼吸道變應性疾病的重要機製之一.
목적:관찰중약복방별민효낭대호흡도변응성모형소서내분비면역공능적영향.방법:이용란청단백건립호흡도변응성소서모형,채용소목정화이홍염색관찰폐조직병리변화,운용류식세포의검측비림파세포중Th1화Th2세포변화,이용매련면역흡부측정방법검측비림파세포배양상청백세포개소4(interleukin-4, IL-4)、백세포개소5(interleukin-5, IL-5)、간우소γ(interferon-gamma, INF-γ)수평급혈청피질동농도.결과:별민능구명현감경모형소서폐조직적염증반응,강저모형소서Th2세포급기세포인자IL-4、IL-5수평,승고Th1세포급기세포인자INF-γ수평,현저억제모형소서내원성피질동수평적하강. 결론:별민대궤체내분비면역계통공능문란적조절작용가능시기치료호흡도변응성질병적중요궤제지일.
Objective: To investigate the effects of Biminne, a traditional Chinese herbal formula, in regulating the endocrine-immune function in mice with airway allergy. Methods: Forty BALB/c mice were randomly divided into four groups: control group, untreated group, Biminne group, and dexamethasone (Dex) group. The murine model of airway allergy was established by intraperitoneal injection sensitization and intranasal challenge with ovalbumin (OVA). On day 28, the mice were sacrificed and the sera were collected. The lung tissue of mice was removed for haematoxylin-eosin staining and the pathological changes were observed. The spleen cells of mice were isolated and the ratio of Th2/Th1 was determined by flow cytometry. The isolated spleen cells were also cultured in 24-well plates in the presence of concanavalin A and the supernatants were collected after 72-hour culture. Levels of interleukin-4 (IL-4), IL-5 and interferon-gamma (IFN-γ) in the supernatants and the corticosterone level in sera were analyzed by enzyme-linked immunosorbent assay.Results: Biminne significantly relieved the inflammatory reaction in the lung tissue of airway allergic mice. Compared with the control group, the Th2 cells in the untreated group increased, while the IFN-γ level decreased and the ratio of Th2/Th1 increased significantly, showing a Th2 response priority. Biminne treatment could down-regulate the Th2 cells and their cytokines IL-4 and IL-5, and up-regulate the Th1 cells and their cytokine IFN-γ. So the ratio of Th2/Th1 decreased significantly, shifting to the control level. In addition, Biminne also inhibited the reduction of the serum corticosterone of sensitized mice. Conclusion: The Chinese herbal formula Biminne can reverse the functional disorder of the endocrine-immune system, which may explain its therapeutic effects on airway allergic disease.
