目的分析lyn基因在Kaposi肉瘤(KS)组织中的表达情况.方法采集新疆地区KS标本及正常皮肤蜡块各17份,新鲜KS及瘤旁正常皮肤组织5份,采用免疫组化Envision二步法和实时PCR技术从蛋白水平及转录水平检测lyn的表达.结果 lyn蛋白呈胞质阳性,17例Kaposi肉瘤组织均呈阳性表达,阳性率为100%;17例正常皮肤组织中2例呈阳性表达,阳性率为11.8%,lyn蛋白在KS中的阳性表达率明显高于正常皮肤组织,差异有统计学意义(x~2=27.57,P＜0.01).5份新鲜KS组织lyn mRNA的相对表达量高于瘤旁正常皮肤组织(t=2.82,P＜0.05).结论 lyn蛋白的异常表达在KS的发展过程中可能发挥了一定的作用.

Objective To analyse the expression of lyn gene in Kaposi's sarcoma(KS).Methods Immunohistochemical method(Envision)was used to detect the protein expression of lyn in tissue samples from 17 patients with KS and 17 normal human controls, and real-time PCR to measure the mRNA expression of lyn in fresh specimens from the tumor and adjacent normal tissues of 5 patients with KS.Results The lyn protein was cytoplasmically expressed in 17(100%)patients with KS and 2(11.8%)normal controls(x~2 =27.57, P ＜ 0.01).The expression intensity of lyn mRNA was significantly higher in fresh tumor tissues than in adjacent normal tissues(t = 2.82, P ＜ 0.05).Conclusion The abnormal expression of lyn may play an important role in the development of KS.
