利用实验室保藏的枯草芽孢杆菌为出发菌株,采用紫外诱变进行选育,得到1株低温碱性蛋白酶的高产菌株,编号为B.Sub3.采用摇瓶液体发酵方式,考察碳源、氮源等营养条件及温度、发酵时间等发酵条件对发酵的影响,结果表明以上因素对低温碱性蛋白酶的生产均有影响.在单因素基础上设计4因素3水平的正交实验,确定B.Sub3的最佳发酵条件为:葡萄糖5 g/L,蛋白胨5 g/L,酵母粉3 g/L,pH=8.0,接种比例为5%,50 mL三角瓶装液量为20 mL,32℃发酵28 h,发酵液的低温蛋白酶活力为2 231 U/mL.

The strains of producing Low-temperature alkaline protease were breeded and the conditions of fermentation were studied.The initial strains were Bacillus subtilis,which was preserved in microbiology laboratory.Strains were breeded by UV mutagenesis,and one strain was obtained,labeling B.Sub 3.The nutrition conditions were examined and the results showed that the composition of the fermentation medium was glucose 5 g/L,peptone 5 g/L,yeast power 3 g/L,pH 8.0.The conditions of fermentation were optimized,and the results showed that the optimal fermentation condition was 5 % inoculation amount in 10 mL of medium filled in 50 mL flasks for 28 h.The production of breeding strains was considerable,and the activity of low-temperature alkaline protease reached 2 231 U/mL at this optimal fermentation condition.
