背景:目前,转基因小鼠越来越多地在脑血管病研究中应用,小鼠的脑缺血再灌注模型是进行该项研究的一种重要模型,因此对该小鼠模型的稳定性和可重复性研究也变得非常必要. 目的:建立重复性强,稳定性好的小鼠大脑中动脉脑缺血模型,为以后对转基因小鼠的研究做好技术准备. 设计:采用配对两组比较的研究方法. 地点和材料:实验在华中科技大学同济医学院附属同济医院器官移植研究所进行,选用 BALB/c和昆明白两种小鼠(均购自同济医科大学动物房) , 5~ 0及 6~ 0两种规格的尼龙丝线,在不同条件下进行试验. 干预:选择 BALB/c和昆明白两种小鼠进行栓塞试验,观察小鼠品系、体质量、线栓规格及术后温度对梗死结果的影响. 主要观察指标:小鼠的梗死体积及神经功能评分. 结果:昆明白小鼠的平均梗死体积为 (12± 4)mm3, BALB/c小鼠的平均梗死体积为 (22± 3 )mm3,两者比较差异有显著性意义( t=16.425,P< 0.01). BALB/c小鼠的神经功能评分与昆明白小鼠比较差异有显著性意义( t=2.005,P< 0.05). 17~ 22 g小鼠匹配 6~ 0的线栓组较 30~ 35 g 的小鼠匹配 6~ 0线栓组梗死体积所占百分比较大( t=14.070 , P< 0.01),神经功能评分也高于后者( t=3.714, P< 0.05). 结论:小鼠品系、小鼠体质量与线栓规格是否匹配、术后温度是否保持恒定均影响小鼠线栓模型的稳定性.必须严格控制上述各种因素以建立稳定的小鼠局灶性脑缺血模型,为脑科学研究提供可靠的技术支持.

BACKGROUND:In recent years,transgenic mice have been widely used in the study of cerebral vascular diseases,for mouse models of focal cerebral ischemia-reperfusion plays a key role.At present, studies on this mouse model remain scarce,especially with Kunmingbai mice that are widely used by Chinese scientist,to render it necessary to study the stablity and reproducibility of this mouse model. OBJECTIVE:To establish the reliable and reproducible mouse models of cerebral ischemia-reperfusion to prepare for further research of transgenic mice. DESIGN:Controlled analysis with paired comparison was carried out. SETTINGS and PARTICIPANTS:The study was conducted in the Organ Transplantation Institute of Tongji Hospital,Tongji Medical College, Huazhong University of Science and Technology.Two strains of mice, BALB/c and Kunming,were used in this study with 5-0 and 6-0 nylon sutures adopted. INTERVENTIONS:The influences of strains and body weight of the mice, the size of the suture for occlusion,as well as the postoperative temperature on the outcomes of infarcts were evaluated after intraluminal occlusion of the middle cerebral artery of the mice. MAIN OUTCOME MEASURES:The infarct volume was observed and neurological deficit scored. RESULTS:The average infarct volume was(12± 4) mm3 for the Kummin mice and(22± 3) mm3 for the BALB/c mice,with significant difference(t=16.425,P< 0.01).There was significant difference between the two strains of mice in the scores of neurological function(t=2.005,P< 0.05).The percentage of infarct volume and the scores of neurological function were higher in the mice with the weight of 17-22 g than in the mice with the weight of 30-35 g,both of which were blocked with 6-0 nylon suture(t=14.070,P< 0.01;t=3.714,P< 0.05). CONCLUSION:Various factors that the influence the stability of the mouse model of focal cerebral ischemia must be strictly controlled.
