目的 了解肝细胞癌(HCC)中RASSF1A、DAPK、FHIT、RIZ1和p73基因的甲基化状况.方法 应用甲基化特异性PCR技术,检测40例HCC和4例正常肝组织中5种基因启动子区域的甲基化状况,并分析每种基因甲基化程度和临床参数之间的关系.结果 正常肝组织中无基因甲基化.癌组织中,RASSF1A、DAPK、FHIT、RIZ1和p73基因的甲基化率分别是:90%、77.5%、70%、30%和5%.相应癌旁组织中,RASSF1A、DAPK、FHIT、RIZ1和p73的甲基化率分别为77.5%、77.5%、62.5%、5%和0%.结论 HCC中多基因的启动子甲基化是一种普遍的事件.

목적 료해간세포암(HCC)중RASSF1A、DAPK、FHIT、RIZ1화p73기인적갑기화상황.방법 응용갑기화특이성PCR기술,검측40례HCC화4례정상간조직중5충기인계동자구역적갑기화상황,병분석매충기인갑기화정도화림상삼수지간적관계.결과 정상간조직중무기인갑기화.암조직중,RASSF1A、DAPK、FHIT、RIZ1화p73기인적갑기화솔분별시:90%、77.5%、70%、30%화5%.상응암방조직중,RASSF1A、DAPK、FHIT、RIZ1화p73적갑기화솔분별위77.5%、77.5%、62.5%、5%화0%.결론 HCC중다기인적계동자갑기화시일충보편적사건.
Objective To study the methylation state of RASSF1A,DAPK,FHIT,RIZ1,and p73 genes in heDatocellular carcinoma.Methods The technique of methylation-specific PCR was adopted to investigate the promoter methylation of these genes in 40 cases of HCC and 4 normal liver tissues.The relationship between the frequency of methylation of the genes and clinical data was analyzed.Results The phenomenon of genes methylation was absent in patients with normal liver.In the samples of HCC,the frequency of methylation in RASSF1A,DAPK,FHIT,RIZ1,and p73 were 90%,77.5%,70%,30% and 5%,respectively.In the non-tumor liver tissues,the frequency of methylation in RASSF1A,DAPK,FHIT,RIZ1,and p73 were detected in 77.5%,77.5%,62.5%,5% and 0%,respectively.Conclusions Methylation of multiple genes is a common event in HCC.
